摘要
目的:观察颐正活脉丸对HRS诱导人脐血管内皮细胞损伤的影响。方法:本研究用高脂兔血清(HRS)培养人脐静脉内皮细胞(EC)造成细胞损伤模型;同时分别加入颐正活脉丸提取液或抗氧化剂维生素E,观察EC中脂质过氧化物(LPO)含量以及EC利用外源性花生四烯酸(AA)合成前列环素(PGI2)的能力。结果:HRS使培养的EC中LPO含量显著增加,AA孵育液中6-Keto-PGF1a含量显著下降,且随着HRS浓度的增加,作用逐渐增强。结论:颐正活脉丸提取液在浓度2.5-20μg/mL时,有与维生素E相似的阻止HRS引起的EC中LPO含量升高和PGI2合成减少的作用。
Objective:To study the yizheng huomaiwan,s effect on injury of human umbilical cord blood vessel endothelial cells.Methods: In this research,cell injury model was made by cultivating human umbilical cord vein endothelial cells with HRS;at the same time,extraction liquid of Yizheng Huomaiwan and antioxidant pills vitamin E were added respectively;the content of LPO in EC and EC's capacity of synthesis PGI2 with exogenous arachidonic acid were observed.Results: HRS has made the content of LPO in EC increase dramatically,and the content of 6-Keto-PGF1a decrease greatly;What's more,with the increase in the concentration of HRS,its function has gradually strenthened.Conclustion: When extraction liquid of Yizheng Huomaiwan has reached such a concentration 2.5~20μg/mL,it has the similar function with vitamin E which can prevent the increase of LPO in EC caused by HRS and the decrease of PGI2 synthesis.
