苯酚-硫酸法测定桃叶鸦葱根中多糖的含量 被引量:4

Determination of Polysaccharides in Scorzonera Sinensis Lipsch,Et Krasch by Phenol-sulfuric Acid Method

摘要 目的:提取桃叶鸦葱根中多糖并测定多糖的含量。方法:采用煎煮法提取桃叶鸦葱根中多糖,采用苯酚-硫酸法测定多糖的含量。结果:煎煮法提取桃叶鸦葱根中多糖的提取率为13.9%。葡萄糖在所试浓度范围(0.03~0.08mg.mL-1)内线性关系良好,r=0.999 2。平均回收率为101.7%,RSD=0.61%。桃叶鸦葱根中多糖平均含量96.64 mg.g-1,RSD=0.10%。结论:苯酚-硫酸法测定桃叶鸦葱根中多糖含量方法稳定可行,简便。 Objective:To extract Polysaccharides from Scorzonera sinensis Lipsch,Et Krasch and determine the content of polysaccharides.Methods:To extract Polysaccharides with decoction method.The contents of the Polysaccharides in Scorzonera sinensis Lipsch,Et Krasch were determined by Phenol-sulfurie acid method.Results:The extraction yield of Polysaccharides was 13.9%,and the standard curve was excellent liner in the concentration range of 0.03~0.08 mg·mL-1 with correlation coefficient of r=0.999 2.The recovery rates was 101.7%,RSD=0.61%.The content of polysacharides from Scorzonera sinensis Lipsch,Et Krasch was 96.64mg·g-1,RSD=0.10%.Conclusion:The method is proved to be stable,feasible and simple.
