红细胞生成素改善血液透析患者血管的Na^+-K^+ATPase α_1亚基表达

摘要 目的探讨红细胞生成素(rHuEPO)治疗慢性肾功能衰竭血液透析患者对血管的Na+-K+ATPaseα1亚基表达的影响。方法选择21例患者应用rHuEPO治疗,17例应用输血治疗。采用RT-PCR方法观察两组治疗后左桡动脉Na+-K+ATPaseα1mRNA表达,同时测定治疗前、后红细胞膜Na+-K+ATPase活性和颈动脉内膜一中层厚度。结果rHuEPO组Na+-K+ATPaseα1mRNA表达上调,Na+-K+ATPase活性升高,而输血组未见改变。结论rHuEPO能改善Na+-K+ATPaseα1mRNA表达。 Objeetive:To investigate the effects of recombinant human erythropoietin(rHuEPO)on the expression of vascular Na + K +ATP ase α 1 subunit in hemodialysis patients. Methods:Anemia was corrected by rHuEPO in 21 patients and by fresh blood transfusion in 17 patients.The targeted hematocrits(Hct)in both groups of patients were 30%.Relatively quantitative RT PCR methods was used to detect Na + K +ATPase α 1 mRNA Levels in left radial artery.Erythrocyte membrane Na + K +ATP ase activty and thickness of intima middle of carotid were examined before and after the Hct correction.Result:The expression Na + K +ATP ase α 1 mRNA was up regulated and activity of erythrocyte membrane Na + K +ATP ase was markedly elevated after rHuEPO treatment.While in transfused patients,no singnificant changs were detected in α 1 mRNA expression Na + K +ATP ase activity. Conclusion:Treatment with rHuEPO improves expression of vascular Na + K +ATP ase α 1 mRNA,whereas transfusion does not.
