摘要
目的:观察单味郁金(Curcumae)水煎剂对四氯化碳(CCl4)所致急性肝损伤小鼠肝细胞免疫调节作用,为开发利用单味郁金水煎剂防治急慢性肝损伤提供实验依据。方法:ICR小鼠48只,雌雄各半,随机分为空白对照组,模型组,阳性药物组以及郁金高、中、低剂量组,每组8只。阳性药组以甘利欣22.5mg·kg-1腹腔注射,每天1次;郁金高、中、低剂量组分别以12、6、3g·kg-1灌胃,每天1次;空白对照组及模型组等体积蒸馏水灌胃,每天1次;连续给药7d,第7天给药后11h;再以0.2%CCl4橄榄油溶液(10mL·kg-1)腹腔注射(空白对照组除外),复制急性肝损伤模型。造模后24h处死,计算肝指数;肝组织HE染色观察病理变化;RT-PCR法检测IL-1β及TNF-a mRNA表达水平;免疫组织化学法观察IL-18蛋白表达水平。结果:与模型组比较,各给药组肝指数明显下降(P<0.05)。HE染色显示:模型组小鼠肝小叶组织结构紊乱、肝窦充血明显、炎细胞浸润明显、肝组织呈局灶性细胞变性或死亡;而各给药组上述病理变化明显减轻。与模型组比较,郁金给药组小鼠肝组织IL-1β及TNF-α mRNA表达水平及IL-18蛋白表达水平均明显下调(P<0.05)。结论:单味郁金水煎剂可能通过抑制肝组织细胞因子IL-1β、IL-18和TNF-α的表达,调节小鼠肝脏免疫功能而减轻CCl4所致的急性肝损伤。
Objective To observe the immunoregulatory effects of Single herb decoction of Curcumae on acute liver injury indued by CCl4in mice,and provide experimental evidence for the development and utilization of single herb decoction of Curcumae in prevention and treatment of acute and chronic liver injury.Methods Forty-eight ICR mice were randomly divided into 6groups:normal group,model group,high,middle and low doses of Curcumae groups,positive drug group (diammonium glycyrrhizinate).The mice in positive drug group were injected intraperitoneal with diammonium glycyrrhizinate(22.5mg·kg-1),once a day;the mice in treatment groups were orally administered with Curcumae(12,6,3g·kg-1),once a day;the mice in normal and model groups were administered with the same volume of saline,once a day;for 7d,1hafter administration at the seventh day,the mice in model and treatment groups were intraperitoneally injected with 0.2% CCl4 (10mL·kg-1);the mice in normal group with the same volume of saline.All the mice were sacrificed 24hafter injection.The liver index was calculated;the pathological changes were examined by HE staining method.The mRNA expressions of IL-1β and TNF-αwere analyzed with RT-PCR.The protein expressions of IL-18was detected with immunohistochemistry staining.Results Compared with model group,the liver indexes in treatment groups were obviously down-regulated (P〈0.05).In model group,there were a lot of inflammatory cells infiltration and necrotic hepatocytes in mice,but the lesions became much slighter in treatment groups.Compared with model group,the mRNA expressions of IL-1βand TNF-αand the protein expression of IL-18in liver of mice were significantly decreased in treatment groups(P〈0.05).Conclusion The Single herb decoction of Curcumae has the protective effects on CCl4-induced acute liver injury by inhibiting the expression of inflammatory factors IL-1β,IL-18,and TNF-αand regulating the immune function of liver in mice.
