摘要
目的研究泽泻水提物的利尿活性及对肾脏髓质水通道蛋白2(AQP2)的调节作用。方法筛选合格利尿大鼠30只,分为生理盐水组、呋塞米组、泽泻100 mg/kg组、泽泻500 mg/kg组、泽泻1 000 mg/kg组,每组6只。观察单次及连续8次给予泽泻水提物后大鼠尿量,尿Na+、尿K+,尿Cl-的水平,使用RT-PCR方法观察肾脏髓质AQP2 mRNA的相对表达含量。结果单次及连续8次给予正常大鼠泽泻水提物均可显著增加尿量,升高尿Na+、尿K+,尿Cl-水平;8次给予正常大鼠泽泻水提物后显著降低大鼠肾脏髓质AQP2 mRNA相对含量表达。结论泽泻水提物具显著利尿活性,其利尿活性与降低肾脏髓质AQP2作用有关。
Objective To evaluate the diuretic effect on the diuretic activity and regulation of aquaporin- 2 (AQP2). Methods Aqueous extract of Rhizorna Alisrnastis(100 mg/kg, 500 nag/ kg, 1 000 mg/kg), normal saline and furosemide groups of SD rats were administrated acgueous extract and their urine output was quantitated and collected 24h and 8days after the doses, then the urinary levels of Na^+ , K^+ , Cl^ were observed. The kidney medulla AQP2 mRNA relative expression was measured with RT-PCR. Results After single dose and 8-day dose of extract of Rhizoma Alisrnastis, the urine output and the urinary levels of Na^+ , K^+ , Cl^- were significantly increased. AQP2 mRNA expression of extract of Rhizorna Alismastis was down - regulated significantly. Conclusion Aqueous extract of Rhizoma Alisrnastis has significant diuretic activity, it has diuretic effect by down- regulating AQP2.
