摘要
目的:探讨银杏叶提取物(EGb)通过诱导血红素加氧酶1(HO-1)的合成,保护小鼠急性肾脏缺血再灌注的损伤及其机制。方法:昆明种小鼠30只,随机分成假手术组、单纯缺血再灌注组、缺血再灌注+EGb组。测定缺血再灌注24h后小鼠血清肌酐与尿素氮值的变化,观察肾脏组织的病理学改变,并通过免疫组织化学方法观察小鼠肾脏血红素加氧酶1的表达。结果:与假手术组相比,单纯缺血再灌注组肾小管上皮细胞呈明显的缺血性改变,血清肌酐与尿素氮值水平均明显升高(P<0.01),肾脏血红素加氧酶1表达明显增强;缺血再灌注+EGb组与单纯缺血再灌注组相比,肾小管上皮细胞缺血性改变减轻,血清肌酐与尿素氮值水平均明显降低(P<0.01),肾脏血红素加氧酶1表达明显增强。结论:EGb通过诱导HO-1的合成,减轻小鼠急性肾缺血再灌注的损伤。
OBJECTIVE To investigate the effects of Ginkgo biloba extract (EGb) on heme oxygenase-1 (HO-1) expression in mice with renal ischemia reperfusion injury(IRI), and to discuss its protection mechanisms. METHODS A model of renal ischemia-reperfusion injury was set up. 30 mice were randomly divided into sham operated group, IRI group, and IRI + EGb group. The changes of the content of serum creatinine and urea value were determined. The renal pathologic changes were ob served. The HO-1 expression in kidney was evaluated by immunohistochemistry. RESULTS As compared with those of sham operated group, some ischemic changes in the renal tubular epithelial cells were observed, the contents of creatinine and urea and the HO-1 expression increased significantly in IRI group, As compared with those of IRI group, some mild ischemic changes in the renal tubular epithelial cells were observed, the contents of creatinine and urea decreased and the HO-1 expression in- creased significantly in IRI + EGb group. CONCLUSION The results indicated that the administration of EGb could increase HO-I expression and alleviate renal ischemia-reperfusion injury in mice.
