摘要
[目的]研究胡杨异形叶发育的细胞与分子调控机制,揭示胡杨的生态适应性。[方法]利用SDS-PAGE及2-DE技术提取及分离同一株胡杨成年树异形叶片的蛋白质,建立适合胡杨叶片蛋白质分离的电泳技术。[结果]发现异形叶蛋白质的表达存在明显差异。由SDS-PAGE图谱可以看出,分子量为57.2,13.2,30.2,23.9,35.9及33.3 kD处的蛋白谱带差异明显。双向电泳图谱表明,胡杨叶蛋白多为分布在pH值5.0~6.5,20~40 kD之间的酸性低分子量蛋白质;共发现73个差异点,其中在锯齿卵圆形叶片中表达丰度上调的为51个点,下调的为22个点。[结论]推测在长期的进化过程中,胡杨叶片基因的调控表达发生改变而长出不同形状的叶片,以更好地适应环境。
[Objective] To study the cellular and molecular mechanism of the development of Populus heteromorphic leaves.[Method] SDS-PAGE and 2-DE technique were used to extract and separate the protein in the heteromorphic leaves of adult Populus euphratica,as well as to establish an electrophoretic technique which was suitable for separating the protein in the leaves of P.euphratica.[Result] Significant difference of protein expression was existed in heteromorphic leaves of P.euphratica.SDS-PAGE pattern showed that the protein with the molecular weight of 57.2,13.2,30.2,23.9,35.9 and 33.3 kD had obvious difference in their protein bands.2-DE map suggested that most of the leaves proteins of P.euphratica were acidic low molecular weight proteins distributed at the pH value of 5.0-6.5 and molecular weight of 20-40 kD;Totally 73 different points were found,51 of which were abundance up and other 22 were abundance down in the serrated ovate leaves.[Conclusion] In the long process of evolution,the regulation of gene expression in P.euphratica leaves was changed so that different shapes of leaves were grown to better adapt to the environment.
