牛轮状病毒二温式PCR检测方法的建立 被引量:5

摘要 根据牛轮状病毒(Bovine rotavrius,BRV)的群特异性基因VP6设计了1对特异引物,建立并优化了能够快速检测BRV的二温式PCR方法。特异性和敏感性试验结果表明,该二温式PCR只对BRV敏感,可扩增获得大小为385 bp的特异性条带,其敏感性为最低能检测到1 pg的BRV RNA;而对其他病毒不敏感。说明所建立的BRV二温式PCR方法是一种快速、特异、敏感的检测方法。 The experiment was conducted to set up and optimize a duel-temperature PCR for rapid detection of bovine rotavirus(BRV).A pair of specific primers was designed according to conserved sequences of specificity to VP6 gene of BRV.The specificity and sensitivity tests showed that this duel-temperature PCR was only sensitive to the BRV for amplifying specific band of 385 bp and it could detect 1 pg of BRV RNA,without any sensitivity to other viruses.The results of our study demonstrated that the optimized duel-temperature PCR was a rapid,specific and sensitive method for detection of BRV.
