摘要
目的:探讨卵巢癌血管生成拟态的临床意义及血管生成拟态与缺氧诱导因子HIF-1α、基质金属蛋白酶MMP-2表达的关系。方法:收集南通大学附属医院2000年1月~2009年5月问经手术切除的卵巢肿瘤组织石蜡标本81例,采用CD34和PAS双重染色方法证实卵巢癌组织中存在VM,探讨卵巢癌组织中VM与卵巢癌组织类型、病理分级、临床分期、肿瘤指标、患者生存时间的相关性,及VM与HIF-1α、MMP-2表达的关系。结果:卵巢癌中出现VM的几率要明显高于交界性卵巢肿瘤;低分化卵巢癌VM发生的几率要高于高分化的卵巢癌;Kaplan—Meier生存分析提示卵巢癌中存在VM的患者生存时间比无VM患者短。VM与HIF—1α蛋白、MMP-2蛋白表达相关。结论:具有血管生成拟态的卵巢癌组织分化低,患者临床预后差;HIF-1α、MMP-2表达与卵巢癌血管生成拟态密切相关。
Objective: To explore the clinical significance of vasculogenic mimicry in patients with ovarian carcinoma and the rela- tionship between vasculogenic mimicry and hypoxia induced factor - 1 α ( HIF - 1 α), matrix metalloproteinase - 2 ( MMP - 2 ) . Methods: 81 paraffin - embedded specimens of ovarian carcinoma after surgical resection in the hospital from January 2000 to May 2009 were collected, CD34 and PAS double staining methods were used to confirm that vasculogenic mimicry existed in ovarian carcinoma, the correlation between vasculogenic mimicry and histological types, pathological grades, clinical stages, tumor markers and survival time was explored, and the re- lationship between vasculogenic mimicry and expressions of HIF - 1 α and MMP2. Results : The rate of vasculogenic mimicry in ovarian carcinoma was significantly higher than that in borderline ovarian carcinoma ; the rate of vasculogenic mimicry in low differentiated ovarian carcinoma was significantly higher than that in high differentiated ovarian carcinoma; Kaplan - Meier survival analysis showed that the survival time in patients with vasculogenic mimicry was shorter than that in patients without vasculogenic mimicry ; vasculogenic mimicry was related to expressions of HIF - 1 α and MMP2. Conclusion : The prognosis of patients with vasculogenic mimicry and low differentiated ovarian carci- noma is poor; the expressions of HIF-1α and MMP2 are related to vasculogenic mimicry.
