副粘病毒表面糖蛋白的表达及其相互作用的研究 被引量:12

摘要 为研究多种副粘病毒融合蛋白(F)及其与血凝素-神经氨酸酶(HN)的相互作用关系,采用T7RNA聚合酶表达系统,将质粒DNA及载有各种F或HN基因的重组质粒DNA于真核细胞内进行表达,然后常规检测表达产物的功能情况。结果显示,每种病毒F单独表达不足以引起细胞融合,只有与同源性HN共同表达时,才能引起细胞融合;与异源性HN共同表达时,则不能引起细胞融合。提示副粘病毒F的细胞融合作用需要HN的辅助,并具有极强的特异性。 Two kinds of glycoproteins, hemagglutininneuraminidase(HN) and fusion(F) proteins, are enfolded in the envelopes of paramyxoviruses and the infected cells. In order to know the mechanism of cell fusion mediated by F protein, the expressions and functions of each F protein, coexpression with HN and their interactions with homologous and heterologous HN proteins were studied. Plasmid DNAs containing different F or HN genes were expressed in eukaryocytes under T7 RNA polymerase expression system and then biological functions of expressed products were assayed with common Gimsa staining. The results showed that when each F itself was expressed, there was no fusion when F and its heterologous HN coexpressed. These results proved that the function of cell fusion mediated by F is very specific. The reasons for the specificity were widely discussed.
