皮瓣缺血再灌注损伤后细胞凋亡基因Bax蛋白表达变化规律的研究

摘要 目的观察皮瓣缺血再灌注损伤后细胞凋亡基因Bax变化的规律,探讨其可能的发病机制。方法在以大鼠腹壁浅血管为蒂的岛状皮瓣缺血再灌注模型上,分别在不同的时间点观察凋亡基因Bax蛋白表达的细胞数和免疫组化的方法,定量测定皮瓣组织中细胞凋亡基因蛋白表达数值的变化规律。结果在实验组中,随着缺血、再灌注时间的延长,Bax蛋白表达急剧增多,至再灌注后6h达到高峰,之后开始缓慢下降,但尚未观察到恢复正常时的时间段;在对照组中,Bax蛋白表达随时问的延长呈缓慢下降的趋势,因此,对照组较实验组各时间段的Bax蛋白表达降低。两组经SPSS11.5统计学分析,差异均有统计学意义(P〈0.05)。结论缺血再灌注等损伤因素可加速细胞的凋亡,本研究为从基因分子水平保护皮瓣组织发生缺血再灌注损伤提供了临床实践的理论依据。 Objective To observe the variational regularity of expression of Bax protein (apoptosis gene) after flap isehemia-reperfusion injury and explore its possible pathogenesis. Methods The island flaps pediceled with abdominal superficial vessels of rats were as the experimental model of isehemia-reperfusion injury. The positive cells of the Bax protein expression were observed at different times and numerical variations of Bax protein expression in the flaps were measured by the method of immunohistochemistry. Results In the experimental group, With the time of ischemia and reperfusion prolonging, the expression of Bax protein increased rapidly, and achieved the peak at 6 hours after reperfusion, after that it decreased slowly; In the control group, we can find the expressive time of Bax protein was longer than that in experimental group and had a tendency of slow descent. The results were inspected by SPSS 11.5 Statistics and proved to have the statistical significance ( P 〈 0.05 ). Conclusion The damage factors, such as ischemia or reperfusion, the genetic and molecular level of protection from the flap tissue isehemia provides a theoretical basis for clinical practice.
