一株海洋微生物产蛋白酶抗肿瘤活性的初步探讨

摘要 研究寄生于沙蚕消化道的嗜麦芽寡养单胞菌D2株外泌胞外蛋白酶(D2蛋白酶)的抗肿瘤活性。选用鼠乳癌细胞EMT6作为靶细胞,经0.2~2000mg/L的D2蛋白酶作用不同时间,采用MTT法对体外培养靶细胞进行细胞增殖抑制率检测,并与阳性对照组(紫杉醇组)和正常细胞对照组(人胚肺细胞组)比较。MTT检测结果显示,D2蛋白酶对鼠乳癌细胞EMT6增殖有抑制作用,且呈现浓度及时间效应关系。检测结果证明D2蛋白酶具有一定的抗肿瘤活性。 The anti-tumor activity of strain protease D2 from digestive tract of Perinereis aibuhitensis Grube was studied.EMT6 cells were cultured in presence of protease at different concentration(0.2~2 000 mg/L) gradients for different times and,then,MTT assay was used to evaluate the cell proliferation inhibition.Similarly,human embryonic lung cells(HELF) were under the same treatment with protease D2 and set up as normal cell control group.The effect of paclitaxel(PTX) on EMT6 cells was regarded as positive control group.MTT test results showed that protease D2 had a certain inhibition effect on proliferation of EMT6 cells,and exhibited an effective relationship between the concentration and time.The test showed that protease D2 had anti-tumor activity in some degree.
