VP28蛋白鸡源抗WSSV卵黄抗体的制备及性能分析 被引量:1

Development of two antibodies in poultry-chicken yolk against white spot syndrome virus(WSSV) using inactivated WSSV and VP28 protein as antigens and their performance analysis

摘要 为了研制鸡源抗对虾白斑综合征病毒(WSSV)卵黄抗体(IgY),利用WSSV毒株及其衣壳蛋白VP28制备成2种不同的疫苗,并分别免疫接种健康蛋鸡群,使鸡群产生不同的IgY,经病毒中和试验证实,2种IgY均有明显的抗WSSV效果,其中WSSV粗提液1∶1000倍稀释时,WSSV灭活苗免疫蛋鸡产生的IgY对南美白对虾的保护率为61.7%,重组VP28疫苗免疫蛋鸡产生的IgY保护率为48.3%;而WSSV粗提液1∶10000倍稀释时,WSSV灭活苗免疫蛋鸡产生的IgY保护率为85.0%,重组VP28疫苗免疫蛋鸡产生的IgY保护率为75.0%。 The present experiment was conducted to develop the immunoglobulin of yolk (IgY) from poultry-chicken against white spot syndrome virus (WSSV). The healthy egg-chickens were immunized with inactivated WSSV or VP28 protein vaccine to induce the formation of two different specific yolk antibodies. Effect of specific antibodies in protecting shrimps (Litopenacus vannamel) against WSSV was analyzed and confirmed by virus neutralization assay. The results showed positive effects of both IgY against the WSSV. The survival rate of shrimp reached 61.7% when infected by IgY derived from WSSV vaccine injected poultry-chicken (1 ×10^-3 dilution of crude extraction solution), while the survival rate of shrimps was found only 4g.3% when infected by IgY derived from recombinant VP28 vaccine injected poultry-chicken. Further, at 1×10^-4 dilution of crude extraction solution of WSSV, the survival rate of shrimps reached 85 and 75%, in case of IgY derived from WSSV and VP28 protein vaccine injected poultry-chickens, respectively.
