摘要
目的青藤碱(Sinomenine,SN)在类风湿关节炎(rheumatoid arthritis,RA)治疗中有较好的临床疗效,对SN相关免疫机制的研究也越来越受到关注。但关于SN在抗原特异性T细胞(antigen special T cell,AST)机制的研究少见报道。文中观察了SN对RA患者环瓜氨酸肽(cyclic citrullinated peptide,CCP)AST体外增殖的影响,探讨SN治疗RA的相关免疫机制。方法体外分离、培养RA患者外周血淋巴细胞,分别设对照组:不加任何干预药物;CCP组:仅加CCP干预,终浓度为20μg/ml;甲氨喋呤(Methotrexate,MTX)组:同时加CCP与MTX干预,终浓度均为20μg/ml;SN低剂量组:同时加CCP与SN干预,SN终浓度为10μg/ml,CCP终浓度为20μg/ml;SN高剂量组:同时加CCP与SN干预,终浓度均为20μg/ml;体外培养72 h后,采用CCK-8法检测细胞增殖吸光度(A)值水平。结果 CCP可体外诱导RA患者外周血淋巴细胞增殖(P<0.01),在此基础上加入不同浓度的SN后,CCP诱导的淋巴细胞增殖被显著抑制(P<0.01),SN高剂量组较之SN低剂量组的抑制程度更为明显(P<0.01)。结论 SN可抑制RA患者CCP-AST的体外活化、增殖,这可能是SN临床治疗RA的免疫机制之一。
Objective Recently,Sinomenine(SN) is widely used for the treatment of the patients with rheumatoid arthritis.But there is few reports about the effect of SN on antigen specific T cell.In this paper,we observed the effect of sinomenine on the proliferation of cyclic citrullinated peptide antigen specific T cell(CCP-AST)from rheumatoid arthritis patients.Methods Peripheral blood lymphocyte(PBL) from RA patients were divided into five groups and cultured respectively in vitro.Blank control group:untreated by drug;CCP group:cells were untreated by CCP(degree of enrichment 20 μg/ml);Methotrexate(MTX)control group:cells were treated by CCP and MTX(each degree of enrichment 20 μg/ml);SN groups:cells were treated by CCP and SN(each degree of enrichment 10ug/ml and 20 μg/ml).The proliferation rate of lymphocytes were measured using by CCK-8 Kit.Results Compared with blank control group: the proliferation rates of CCP group were significantly higher(P0.01).When cells were treated by SN,the proliferation rate of SN group were significantly inhibited(P0.01).The proliferation rate of high degree of enrichment 20 μg/ml significant proliferated were significantly higher than the low degree of enrichment 10μg/ml(P0.01).Conclusion SN can inhibit the proliferation of the cyclic-citrullinated peptide antigen specific T cell from the peripheral blood lymphocyte of RA patients,which may have important role on treatment of RA.
