摘要
研究刺五加叶水溶性多糖结构信息和免疫活性。采用热水提取、乙醇沉淀、Sevag法脱蛋白后得到刺五加粗多糖,粗多糖经EDAE-Sepharose Fast Flow离子交换柱和Sephadex G-75凝胶过滤柱2次纯化,得到均一多糖组分ASP-2-1;ASP-2-1经三氟乙酸水解、1-苯基-3-甲基-5-吡唑啉酮(PMP)衍生化、毛细管区带电泳法(CZE)测定其单糖组成;高效凝胶渗透色谱法测定其纯度和分子量;体外脾淋巴细胞增值试验测定其免疫活性。从刺五加叶中分离纯化得到的均一多糖组分SPA-2-1主要由葡萄糖、阿拉伯糖、半乳糖、果糖、鼠李糖、木糖等6种单糖组成,摩尔比为27.33:8.92:3.03:1:7.59:20.51,其分子量为13.6 KD;小鼠脾淋巴细胞增殖试验表明,SPA-2-1可显著促进脾淋巴细胞的增殖,刺五加叶多糖SPA-2-1具有较强的体外免疫活性。
Structural information and immunological activity water-soluble polysaccharide from leaves of Acanthopanacis Sentieosi were investigated. Crude polysaceharide was obtained from leaves ofA. Senticosi after removing protein by hot water extraction, ethanol precipitation and Sevag. A homogeneous polysaccharide, named ASP-2-1, was separated and purified from the crude polysaccharide through EDAE-Sepharose Fast Flow Ion-exchange chromatograph and Sephadex G-75 gel filtration chromatography. The monosaccharide components of ASP-2-1 were determined by capillary zone electrophoresis (CZE) after trifluoroacetolysis and l-phenyl-3-methyl-5-pyrazolone (PMP) derivatization in tendorn order. The purity and molecular weight of ASP-2-1 were determined by HPGPC. The immunological activity of ASP-2-1 was menstruated with spleen lymphocytes of mouse in vitro experiment. The ASP-2-1 whose molecular weight was 13.6 KD was mainly composed of glucose, arabinose, galactose, fructose, rhamnose and xylose with the mole ratio of 27.33:8.92: 3.03:1:7.59:20.51. ASP-2-1 showed proliferating effect on splenic lymphocyte markedly. The ASP-2-1 from leaves of A. Senticosi had strong immunological activity in vitro.
