摘要
目的:研究知柏地黄汤对解脲支原体(Uu)感染大鼠睾丸组织IL-2、TNF-α表达水平的影响,探讨该方防治Uu感染所致的男性不育症的部分免疫学机制。方法:经膀胱接种建立大鼠睾丸组织感染解脲支原体的动物模型。实验设知柏地黄汤治疗组、美满霉素治疗组、模型对照组和假手术组。假手术组、模型对照组灌胃给生理盐水,治疗组灌胃给相应药物,连续给药21d后取标本检测指标。免疫组织化学染色法和Western blotting技术检测睾丸组织IL-2、TNF-α蛋白表达水平,RT-PCR技术检测睾丸组织IL-2、TNF-α基因表达水平。结果:Uu感染大鼠后,睾丸组织IL-2的蛋白表达和基因表达水平降低,TNF-α的蛋白表达和基因表达水平升高,与假手术组比较,差异显著(P<0.01);知柏地黄汤组睾丸组织IL-2蛋白表达和基因表达水平明显升高,TNF-α的蛋白表达和基因表达水平明显降低,与模型对照组比较,差异显著(P<0.01)。结论:知柏地黄汤能通过影响Uu感染大鼠睾丸组织IL-2、TNF-α的表达水平而发挥抗感染作用。
Objective: In order to know the immune mechanism of Zhibai Dihuang Decoction(ZHBDHD) in treating male infertility by ureaplasma urealyticum(Uu),we performed the research for the effects of ZHBDHD on the expression of IL-2 TNF-α in the rats infected by Uu.Methods: Rats model was prepared by inoculating Uu in the bladder of rats.These rats were randomly divided into four groups: ZHBDHD group,minocycline group,model group and false operation group.The rats of model group and false operation group were taken NS by garage,twice a day for continuing 21 days;ZHBDHD group were taken ZHBDHD and minocycline group taken minocycline by garage as the other two groups.Stop water and food for 8 hours after the last garage,taken samples for determining the contents of IL-2,TNF-α in testis by immunohistochemistry and western bloting;determining mRNA of IL-2,TNF-αin testis with RT-PCR.Result: Compared with false operation group,the expressions of IL-2 in testis were obviously decreased after infected by Uu.However,TNF-α in testis were found obviously increasing after rats infected by Uu comparing with false operation rats;On the contrary,ZHBDHD could increase the expression of IL-2,inhibit the expression of TNF-α.The results were same when ZHBDHD group was compared with minocycline group.Conclusion: ZHBDHD can exert anti-inflammatory by effecting the expression of IL-2 and TNF-α in the rats infected by Uu.
