在 p H 1.8的 K H2 P O4 H3 P O4 缓冲介 质中,蛋白质与酸性 品红结合 使染料的 吸光度下 降。蛋白质 浓度 C 在0. 40—4.00m g · L- 1 和 8.00—40.00m g· L- 1 范 围内,吸 光度的 降低( Δ A) 与蛋白 质浓 度( C)呈 线性 关系,回 归方 程分 别为 Δ A= 0.0140 C+ 0. 0534 和 Δ A= 0. 0109 C+ 0.0895, 相关 系数 r 依次 为 0. 9965和 0.9993,方法用于奶粉和麦片中蛋白质的测定,结果满意。
The colour of fuchsine acid dye is reduced by the interaction of protein with fuchsine acid in buffer solution of KH 2PO 4 H 3PO 4(pH1.8).The linear relationship holds between the reduced absorptivity of dye and the protein concentration ranging from 0.40—4.00mg·L -1 and 8.00? ?0.00 mg·L -1 and the regression equations are Δ A =0.0534+0.0140 C (mg·L -1 ) and Δ A =0.0895+ 0.0109 C with the correlation coefficient to be 0.9965 and 0.9993,respectively.The proposed method has been applied to determine protein in milk powder and oatmeal with satisfactory results.
