摘要
目的观察肝素对内皮细胞(endotheliumcel,EC)增殖与内皮源性收缩因子释放的影响。方法(1)采用人脐静脉EC培养技术,绘制EC生长曲线;(2)用扫描电镜观察大鼠颈总动脉血管吻合口EC的覆盖率;(3)采用牛主动脉EC培养及离体血管舒缩功能实验技术,观察肝素对血管的直接解痉作用及对培养EC释放收缩因子的影响。结果(1)即使浓度高达500μg/ml的肝素对培养EC的增殖也无作用,但能明显增强内皮细胞生长因子(ECGF)对EC增殖的促进作用。肝素加ECGF组的EC数比ECGF组增长了40%~74%;(2)在血管吻合术后24小时,对照组血管吻合口未见EC移行,肝素组EC覆盖了近25%。术后48小时,肝素组的EC覆盖率是对照组的2倍;(3)肝素无直接的血管解痉作用。但可使缺氧和低温的条件培养液的血管收缩作用分别减弱55%和59%。结论肝素对培养EC的增殖及离体的收缩血管无直接作用,但能辅助ECGF促进EC增殖,并抑制EC合成与释放内皮源性收缩因子。
Objectives (1)To

Methods (1)Adopting culturing technique of human umbilical vein endothelial cell,growth
curve of endothelial cell was drawed (2)using scanning electron microscopy technique,the
endothelial coving rates of anastomotic sites of rat common carotid arteries were
assayed (3)The techniques of bovin aortic endothelial cell culture and vasomotorial
functional test of isolated blood vessel,the influence of heparin on the relaxing directly
effect of blood vessel and the releace of contractile factors by cultured endothelium was
observed Results (1)Heparin at concentration as high as 500 μg/ml did not support
replication,but could enhanced endothelial cell growth factor(ECGF)promoted cultured
endothelium growth The numbers of endothelium in heparin and ECGF group increased
40%~74% relative to ECGF group (2)The migration of endothelium at anastomotic sites
in control group was not obser ved ,but endothelium coving in 25% of anastomotic
site in heparin group at 24 hours after the anastomosis of blood vessel The endothelial
coving rates in heparin group was double relative to control group at 48 hours
postoperatively (3)Heparin could not relax directly contractile blood vessel,but could
reduced respectively the conditioned medium form anoxic and cold group induced
constrictions to 55% and 59% Conclusion Heparin has not directly effect on the
proliferation of cultured endothelial cell and contractile blood vessel,but could enhanced
ECGF promoted endothelial cell growth and inhibit release of endothelium derived
contractile factors
