p-PKB和NF-κBp65在宫颈癌组织中的表达及意义 被引量:1

The Expressions of p-PKB and NF-κBp65 in Cervical Carcinoma Tissues and Their Significances

摘要 目的探讨宫颈癌组织中磷酸化蛋白激酶B(p-PKB)和核转录因子-κB p65(NF-κBp65)蛋白的表达及意义。方法采用免疫组化S-P法检测51例宫颈癌组织、36例宫颈上皮内瘤变(CIN)组织和20例正常宫颈组织中p-PKB和NF-κBp65蛋白的表达,并分析两者与宫颈癌临床病理参数的关系。结果 p-PKB蛋白在宫颈癌组织中的阳性表达率(74.5%)明显高于CIN组(52.8%)和正常宫颈组(15.0%)(P<0.05),p-PKB蛋白的表达随宫颈癌分化程度下降、临床分期升高而升高(P<0.05),在无淋巴结转移的宫颈癌组织中的表达明显低于有淋巴结转移者(P<0.05)。NF-κBp65蛋白在宫颈癌组织中的阳性表达率(49.0%)明显高于CIN组(30.6%)和正常宫颈组(16.7%)(P<0.05),NF-κBp65蛋白的表达随宫颈癌分化程度下降、临床分期升高而升高(P<0.05),在无淋巴结转移的宫颈癌组织中的表达明显低于有淋巴结转移者(P<0.05)。结论 p-PKB、NF-κBp65蛋白在宫颈癌组织中过表达与宫颈癌的发生发展有关,p-PKB蛋白可能通过作用于其下游NF-κBp65蛋白,促进宫颈癌的恶性转化。 Objective To investigate the expressions of phosphorylated protein kinase B(p-PKB) and NF-κBp65 protein in cervical carcinoma tissues and their significances.Methods The expressions of p-PKB and NF-κBp65 protein were measured by immunohistochemical staining in 51 cases of cervical carcinoma,36 cases of cervical intraepithelial neoplasia(CIN),and 20 cases of normal cervical tissues.The relationship between p-PKB,NF-κBp65 protein and clinicopathological factors in cervical carcinoma were analyzed.Results The positive rate of p-PKB protein in the cervical carcinoma tissues(74.5%) was remarkably higher than those in the CIN tissues(52.8%)and the normal cervical tissues(15.0%)(P0.05).The positive rate of p-PKB protein in the cervical carcinoma tissues became higher when the tumor differentiation was poor,the clinical stages were late and the lymph metastases were positive(P0.05).The positive rate of NF-κ Bp65 protein in the cervical carcinoma tissues(49.0%) was remarkably higher than those in the CIN tissues(30.6%) and the normal cervical tissues(16.7%)(P0.05).The positive rate of NF-κBp65 protein in the cervical carcinoma became higher when the tumor differentiation was poor,the clinical stages were late and the lymph metastases were positive(P0.05).Conclusion The overexpressions of p-PKB and NF-κBp65 in cervical carcinoma correlate with the tumorigenesis and progression of cervical carcinoma.p-PKB may promote the malignant transformation of cervical carcinoma through up-regulation of NF-κBp65 protein.
