Investigation of protein staining protocols of SDS-polyacrylamide gel electrophoresis

摘要 目的建立一种操作简便、快速、灵敏度高、无毒的蛋白质电泳染色方法。方法利用考马斯亮蓝G-250(CBB G-250)在SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)后对蛋白质条带进行染色,对不同染色条件进行探讨,并与考马斯亮蓝R-250(CBB R-250)染色法进行比较。结果 CBB G-250染色法试剂无毒,背景浅,经改良其灵敏度与CBB R-250染色法相当,达到0.1~0.5μg。结论 CBB G-250染色方法简单经济,灵敏度能够满足一般要求,适用于蛋白质电泳结果的快速分析。 Purpose To establish a simple,rapid,highly sensitive,non-toxic protein electrophoresis staining protocol.Methods By utilizing Coomassie Brilliant Blue G-250(CBB G-250) to stain the pro-tein bands by SDS-polyacrylamide gel electrophoresis(SDS-PAGE),the different dyeing conditions were explored and compared to the staining of Coomassie Brilliant Blue R-250(CBB R-250).Results CBB G-250 staining protocol presents non-toxic,light background and comparable sensitivity after optimization toCBB R-250 staining,which is reaching 0.1-0.5 μg.Conclusion CBB G-250 staining protocol is simpleand economic.Its sensitivity could meet the general requirements,and is applicable to the rapid analysis of protein electrophoresis.
