环孢霉素A对缺血再灌注心肌的保护作用与CD4^+ T细胞的关系 被引量:2

摘要 目的观察环孢霉素A对缺血再灌注心肌的影响,并探讨其与CD4+T细胞之间的关系。方法将32只SD大鼠随机分为4组(n=8):假手术组(Sham)、缺血再灌注(I/R)模型组、环孢霉素(CsA)组、CsA+5-羟基葵酸盐(5-HD)组。CsA、CsA+5-HD组给予CsA 15 mg/kg及1 mL生理盐水腹腔注射,Sham组和I/R组注入相同剂量的生理盐水,连续5 d,CsA+5-HD组于术前30 min加用5-HD腹腔注射(10 mg/kg)。阻断冠状动脉左前降支30 min,复灌24 h,免疫荧光染色计数浸润的CD4+和CD3+T淋巴细胞,RT-PCR及ELISA检测钙调神经磷酸酶(CaN)的表达,组织病理学观察心肌细胞的损伤情况。结果与I/R组相比,CsA组及CsA+5-HD组CD4+T细胞表达明显减少(P<0.05),CaN表达降低(P<0.05),而病理损伤程度减轻。结论 CsA被取消作为线粒体通透性转化孔抑制剂作用后仍能对缺血再灌注心肌产生保护作用,其作用可能与抑制CD4+T细胞的激活有关。 Objective To study the protective effect of cyclosporine A on myocardial ischemia-reperfusion injury(MIRI) in rats,and to investigate the correlation between the protective effects and CD4+ T cells.WEN Bing,YANG Shuang-qiang.Department of Cardiothoracic,The First Affilated Hospital of Chongqing Medical University,Chongqing 400016,ChinaMethods Thirty-two rats were randomly divided into four groups(n=8): sham operation group,ischemic-reperfusion(I/R) group,CsA group and CsA+5-HD group.Intraperitoneal injection of CsA(15 mg/kg) and saline(1 mL) was performed in CsA and CsA+5-HD groups for 5 days,while same volume of saline was given in sham operation and I/R group.In CsA+5-HD group,5-Hydroxydecanoic acid sodium salt(5-HD) was injected 30 minutes before operation.Rats were subjected to left anterior descending coronary artery occlusion for 30 min and subsequent reperfusion for 24 hours.The CD4+ and CD3+T cell counts were examed with immunoflurorescent staining.The expression of CaN mRNA and enzyme were detected by RT-PCR and ELISA.Results The CD4+ and CD3+ T cell count and expression of CaN were significantly reduced in CsA and CsA+5-HD groups,comparing with those in I/R group(P0.05).Correspondingly,milder pathological injury was observed in CsA and CsA+5-HD groups than that in I/R group.Conclusion Cyclosporine A shows protective effect on rat MIRI even with mitochondrial permeability pore inhibitors,probably via inhibition of CD4+T cell.
