滋阴活血方药和尿酸盐对糖尿病大鼠牙周组织的影响 被引量:1

Effect of NYPBR and urate on the structure of periodontal tissue of diabetic rats

摘要 目的对比观察滋阴活血方药(nourishing yin and promoting blood-flow recipe,NYPBR)和尿酸盐(urate)对糖尿病大鼠牙周组织结构的影响。方法选用6月龄雄性Wistar大鼠38只随机分为4组,对照组8只、糖尿病组、中药组和西药组各10只。建立大鼠糖尿病模型,中药组给予NYPBR,西药组给予urate。给药后12周处死大鼠,取上颌骨标本,行组织学及骨计量学观察。结果①组织学观察:对照组牙周组织无明显变化;糖尿病组大鼠12周时牙龈上皮和固有层中度炎细胞浸润,牙龈和牙周膜内毛细血管数目增多、扩张充血,纤维排列紊乱和断裂,骨吸收陷窝多,陷窝内见破骨细胞,牙槽嵴顶无吸收;中药组、西药组牙周组织结构无明显异常,结构与对照组相似。②骨计量学观察:12周时大鼠牙周结缔组织附着丧失量和牙槽骨嵴高度丧失量各组间无差异(P>0.05)。对照组、中药组、西药组成骨细胞数和破骨细胞数无明显差异(P>0.05),成骨细胞数明显高于糖尿病组(P<0.05),破骨细胞数明显低于糖尿病组(P<0.05)。结论 NYPBR和urate可有效预防糖尿病造成的牙周组织受损。 Objective To observe the effect of nourishing yin and promoting blood-flow recipe(NYPBR) and urate on the structure of periodontal tissue of diabetic rats. Methods A total of 38 six-month-old male Wistar rats were randomly divided into four groups,with 8 in the control group,and 10 in DM group,the NYPBR group and the urate group.After DM models were made,NYPBR and urate were administered respectively to the rats in the respective groups.After 12 weeks,the rats were killed and the maxilla was collected for histological examination and bone histomorphometry study. Results ①Histological observation: No pathological changes occurred in periodontal tissue of the control group.In the DM group,inflammatory cells infiltration in gingiva epithelium and lamina propria were observed.The number of blood capillary increased,with vessel lumen dilated and hyperemic.Collagen fibrils were in disorder and fractured.Osteoclasts were found in the increased osseous lacunas under the alveolar bone.However,there was no absorption on alveolar ridge crest.The structure of the NYPBR group and urate group was similar to that of the control group.②Bone histomorphometry observation: There was no significant difference in the loss of connective tissue attachment and alveolar bone height among the four groups(P0.05).There was no significant difference in osteoblasts and osteoclasts number among the control group,the NYPBR group,and the urate group(P0.05).But the osteoblasts number of the three groups above was higher than that of the DM group(P0.05),and the osteoclasts number was lower than that of the DM group(P0.05). Conclusion Both NYPBR and urate can prevent the injury of periodontal tissue caused by DM.
