摘要
目的探讨水蛭素对体外培养的人增生性瘢痕成纤维细胞(HSFb)凋亡的作用。方法分离培养HSFb取4~7代细胞实验。分别采用免疫细胞化法及Western blot法检测不同浓度水蛭(0 ml、0.25 ml、1.00 ml)对HSFb、Bax、Bcl-2基因表达的影响。结果水蛭素对HSFb的增殖有抑制作用;免疫组化实验及Western blot法检测均表明:水蛭素能增加Bax表达,抑制Bcl-2表达,和对照组比较差别有统计学意义(P<0.05)。结论实验浓度水蛭素对HSFb有显著的抑制作用。
Objective To explore the effect of hirudin on the apoptosis of the human hypertrophic scar fibroblast(HSFb)cultivated in vitro.Methods Experiment of isolate cultivating HSFb and cells after 4-7 generations were used in subsequent experiments.The expression of Bax and Bcl-2 in HSFb were analyzed by Western blot.Results Hirudin had an inhibitive effect on the proliferation of HSFb;Immunohistochemistry and Western blot results showed that hirudin increased the expression of Bax,but inhibited the expression of Bcl-2,compared with the control group,the difference was statistically significant(P0.05).Conclusion Hirudin of experimental concentration has significant inhibitory effects on HSFb.
