摘要
目的:研究3-甲氧基-4-O-(2'-亚甲基-3'5'6'-三甲基吡嗪基)肉桂酸(MC-002)抗高血压治疗对大鼠脑卒中的保护作用。方法:采用双肾一夹(2K1C)法复制RHRSP(stroke prone renovascular hypertensive rats)模型引发肾性高血压,高钠饮水进一步加速脑卒中形成。观察MC-002各剂量组静脉注射给药后对脑卒中大鼠神经症状、体重、血压、脑组织病理切片的影响。结果:MC-002能明显改善大鼠的神经症状,显著降低大鼠的血压(P<0.01 vs模型组);30、18 mg/kg剂量组能显著抑制脑卒中大鼠的体重下降(P<0.01vs模型组);脑组织病理切片显示,治疗组神经细胞形态和数目均得到显著改善。结论:MC-002作为一种新型的Ca2+拮抗剂和自由基清除剂,抗高血压治疗对大鼠脑卒中具有明显的保护作用。
AIM: To investgate the protection effects of 3-methoxy-4-O-(2'-methyl radical-3'5'6'-trimethyl pyrazinyl) cinnamic acid(MC-002) on anti-stroke in rats by antihypertensive therapy. METHODS: RHRSP(stroke prone renovascular hypertensive rats) model was reproduced to initiate 2K1C renal hypertension,and the stroke was accelerated by high sodium intake.The effect of MC-002 at different doses on rats' neurological symptom,body weight,blood pressure and pathological section of brain tissues were examined after the administration by i.v.injections.RESULTS:MC-002 can significantly improve the rats' neurological symptom and decrease blood pressure(P0.01);MC-002(30 and 18 mg/kg) can markedly inhibit the dropping of rats' bodyweight(P0.01);the pathological observations showed that neuronal cells shape and number were obviously improved after treated with MC-002.CONCLUSION: As a new Ca2+ antagonist and scavenger of free radicals,MC-002 has notable effects of stroke protection in rats by antihypertensive therapy.
