摘要
目的:观察桃红四物汤对寒凝血瘀模型大鼠卵巢血管舒-缩因子及卵巢功能的影响。方法:将大鼠置于0℃1℃冰水中造成寒凝血瘀大鼠模型,采用阴道涂片检测大鼠动情周期;采用放射免疫法测定血清E2、P、T水平;剖取卵巢,制成10%的匀浆,测定ET-1、NO、Ang-2、CGRP活性。结果:模型组大鼠动情周期延长,血清E2、P、T明显下降,卵巢组织匀浆ET-1、Ang-2活性升高,NO、CGRP活性降低,与正常组比较有统计学意义;桃红四物汤组动情周期缩短,血清E2、P、T明显升高,卵巢组织匀浆ET-1、Ang-2活性降低,NO、CGRP活性升高,与模型组比较有统计学意义。结论:桃红四物汤通过调节血管舒缩功能,改善生殖激素水平治疗寒凝血瘀型妇科疾病。
Objective: To observe the effect of Taohongsiwu Decoction on model rat of blood stasis due to accumulation of cold in vasorelaxation-vasoconstriction factors and in ovarian function.Methods: The rats model of blood stasis due to accumulation of cold were induced by keeping in water at 01℃.The estrous cycle was detected by vaginal smear.The level of serum E 2,P,T were measured by radioimmunoassay.Rats’ ovaries were prepared for the solution of 10% homogenate to measure ET-1、NO、Ang-2、CGRP.Results: Compared with the normal group,The estrous cycle in model group was significantly prolonged and the level of serum E 2、P、T and the activity of NO、CGRP in ovary homogenate were significantly decreased.The concentrations of ET-1,Ang-2 in ovary homogenate were increased.After administration with Taohongsiwu Decoction,The estrous cycle was significantly decurtated and the level of serum E 2、P were increased.The activity of ET-1,Ang-2 in ovary homogenate were decreased.Meanwhile the activity of NO、CGRP in ovary homogenate were increased.There was positive correlation between P and NO,CGRP in ovary homogenate,a negative correlation between P and ET-1、Ang-2 in ovary homogenate.There was negative correlation between E 2 and ET-1 in ovary homogenate.Conclusion: The therapeutical effect of Taohongsiwu Decoction on gynecological desease may act through regulating vasomotor function and improving the level of reproductive hormone.
