摘要
以乌拉尔甘草和光果甘草诱导的毛状根为材料,采用固体和液体不同培养方式提供种源,实验分析了毛状根种子来源、培养方式、培养温度、转速和通气量等因素对甘草毛状根生长的影响,研究了甘草毛状根的摇瓶培养,5L发酵培养,并完成了10L搅拌式培养条件的优化。结果表明在毛状根接种量为1‰~3‰、培养温度为25℃、转速为40r/min、通气量为0.5vvm条件下,10L搅拌式培养的甘草毛状根的生物量在60d可增殖55~60倍,不低于摇瓶培养。高效液相法检测出甘草毛状根中甘草酸含量为0.1%。
Some factors that affected the hairy roots growth of Glycyrrhiza uralensis fisch and Glycyrrhiza inflata bat were studied,including the source of fermentation seed,the temperature,the rotate speed and the dissolved oxygen in medium.According to the parameter data of fermentation in 5L culture and 0.5L flask cultivation,the technology of 10L stirred bioreactor was established.The results showed that:using 1‰~3‰ inoculums size,25℃,40r/min and rotated speed 0.5vvm applied air,the growth rate of the 60 days hairy roots obtained from 10L stirred bioreactor was 55~60 no less than those cultured in flask.The glycyrrhizinic acid was detected in hairy roots of Glycyrrhiza uralensis fisch by HPLC whose content was 0.1% dry weight.
