摘要
探讨粉防己碱对培养的U937细胞中花生四烯酸瀑布系统产生前列腺素的影响。以U937细胞株为模型,加入内毒素(LPS)处理,以及粉防己碱预处理后再加入LPS处理,观察Ⅱ-型磷脂酶A2(Ⅱ—PLA2)、胞液型磷脂酶A2-(cPLA2)和环氧合酶-2(COX—2)以及前列腺素E2(PGE2)的变化。结果发现LPS可刺激U937细胞中Ⅱ-PLA2、cPLA2和COX—2以及PGE2的产生增加,粉防己碱则可不同程度地抑制它们的增高。粉防己碱具有降低内毒素引发的Ⅱ—PLA2、cPLA2和COX—2以及PGE2产生的作用。
The purpose of thin study was to investigate the inhibitory effects of tetrandrine on the production ofII-phospholipase A_2 (II - PLA_2), cytosolic phospholipase A_2 (cPLA_2), cyclo - oxygenase 2, COX - 2 (orprostaglandin synthase 2) and prostaglandin E_2(PGE_2) in U937 cells induced by lipopolysacoharide(LPS). Cultured U937 cells were separately treated by adding LPS and tetrandrine. II-PLA_2 activity and the levels of cPLA_2,COX-2 and PGE_2 in U937 cells were analyzed by radioactive assay or ELISA. It was found that LPS couldincrease the activity of II-PLA_2 and stimulate U937 cells to produce more cPLA_2, COX-2 and PGE_2.Tetrandrine could inhibit the increase of these indicators induced by LPS to different degrees. Our findingssuggest that tetrandrine plays the role of anti- inflammation through inhibiting the production of phospholipaseA_2, prostaglandin synthase 2 and prostaglandin by U937 cells.
