不同疗程的灯盏细辛对大鼠急性脑缺血恢复期的保护作用 被引量:3

摘要 目的观察不同疗程的灯盏细辛对急性脑缺血恢复期的保护作用。方法采用改良的Zealonga法建立大鼠大脑中动脉局灶缺血/再灌注梗死(middle cerebral artery occlusion/reperfusion,MCAO)模型。将大鼠随机分为MCAO模型组、短期给药组(腹腔内注射灯盏细辛7 d)、较长期给药组(腹腔内注射灯盏细辛28 d)和正常对照组4组。分别于MCAO术后1 h、1周、2周、4周末进行神经功能评分,分别计算1周、2周、4周末与1h的差值。4周末进行脑梗死相对体积比(%)测定和电镜观察梗死边缘神经元结构。结果各组神经行为学评分差值及脑梗死体积间比较有统计学差异(P<0.05),且较长期给药组效果优于短期给药组(P<0.05)。电镜下观察显示较长期给药组大鼠脑组织病理学改变较轻。结论较长疗程灯盏细辛治疗对脑缺血恢复期的神经保护作用较短期治疗明显。 Objective To observe the protection of breviscapine on acute cerebral ischemia during restoration period in rats. Methods The middle cerebral artery occlusion (MCAO) model was made in rats. 60 rats were randomly divided into 4 groups: (1) MCAO model group, rats were treated with 2.7 mL / kg saline, injected intraperitoneally for 7 days; (2) Short-term treatment group, rats were treated with 2.7 mL / kg breviseapine, injected intraperitoneally for 7 days; (3) Long-term treatment group, rats were treated with 2.7 mL / kg breviscapine, injected intraperitoneally for 28 days; (4) Sham group, rats were treated with 2.7 mL / kg saline, injected intraperitoneally for 7 days. The neurological deficit scores were recorded I h, 14 d, 28 d post MCAO; and the scores between 1 h and any of other different time points were compared separately. Twentyeight days after MCAO, the infarct volume ratio and pathological changes were explored. Results (1) The neurological deficit scores were significantly decreased in long term treatment group, short-term treatment group, and MCAO model group as compared with sham group (P〈0.05); and the infarct volume ratio was also different among the animal groups (P〈0.05). (2) Above changes were more prominent in long-term treatment group than short-term treatment group and MCAO model group (P〈0.05) ; (3) The pathological changes were more minimal in the long-term treatment group than other groups under electron microscopy. Conclusions The long term treatment with breviscapine have better neuroprotective effects after cerebral ischemia.
