摘要
目的:观察半边莲生物碱对高血压鼠脑基底动脉血管平滑肌细胞增殖的作用.方法:分别使用50、100和150mg/L不同剂量半边莲生物碱孵育高血压鼠脑基底动脉平滑肌细胞,以细胞计数试剂盒计数各组细胞个数、[3H]-TdR掺入检测各组DNA的合成量作为细胞增殖的指标.结果:各组细胞数量下降;各组血管平滑肌细胞DNA的合成量均下降,以100mg/L组下降最为明显.结论:半边莲生物碱可抑制高血压所诱导的脑基底动脉血管平滑肌细胞增殖,提示半边莲生物碱可用于高血压诱发的脑血管重构.
Objective: To investigate the erfects of lobeIia chinensis lour alkaloids (LCLA) on smooth muscle cell proliferation of cerebral basilar artery in hypertensive rats. Methods: The smooth muscle cells of the cerebral basilar artery in hypertensive rats were incubated by 50, 100 and 150 mg/L LCLA in each group. Then, to observe the cell proliferation in each group, the cell number was quantified by cell counting kit and DNA synthesis was measured by ^3H-TdR incorporation. Resuits: Compared with the control group, the cell number of each group was markedly declined; DNA synthesis of smooth muscle cells in each group was obviously descended. The drop of 100 mg/L group was most famous among three groups. Conclusion : LCLA may significantly inhibit the hypertension-induced proliferation of smooth muscle cells in the cerebral basilar artery, suggesting that LCLA may prevent and treat hypertension-induced cerebrovascular remodeling.
