摘要
目的:探索糖皮质激素性骨质疏松症的分子机理之一可能由于脂类代谢异常所致,研究补肾中药对糖皮质激素性骨质疏松症的防治作用。方法:采用肌肉注射地塞米松(2.5mg/kg)和喂食低钙饲料的方法建立大鼠骨质疏松模型。将清洁级WISTAR大鼠(143只,雌雄各半)随机分成6组,分别是:A组:正常组;B组:模型空白对照组;C组:补肾中药组;D组:补中益气颗粒对照组;E组:血府逐瘀胶囊对照组;F组:骨疏康颗粒对照组。连续给药9周后,双能X线骨密度仪测定其股骨骨密度;Western blotting测定骨组织脂肪细胞型脂肪酸结合蛋白(A-FABP)表达;自动生化分析仪测定各组血清高密度脂蛋白胆固醇(HDL-C)含量。结果:①与正常组比较,模空组大鼠股骨骨密度显著降低(P<0.01),股骨A-FABP表达显著增强(P<0.01),血清HDL-C含量显著降低(P<0.01);②与模空组比较,补肾中药组股骨骨密度显著增加(P<0.01),股骨A-FABP表达显著降低(P<0.01),血清HDL-C含量显著升高(P<0.01)。结论:糖皮质激素性骨质疏松症病机与A-FABP升高、HDL-C降低有关,提示具有脂类代谢异常。采用补肾填精益髓的治法(鹿茸、牡蛎、淫羊藿3味中药组方)可以有效地防治糖皮质激素性骨质疏松,其疗效机制与调整脂类代谢有关。
Objective:To explore one molecular mechanism of glucocorticoid induced osteoporosis(GIO)may be the abnormal lipid metabolism and to study the effect of tonifying kidney Chinese herbs on GIO.Method:The GIO model of rats was established by injecting dexamethasone and feeding low-calcium feed.The 143 WISTAR rats(half male and female)were divided randomly into six groups.They are normal group A,model group B,tonifying kidney group C,tonify spleen Chinese herbs group D,Xuefu zuyu group(a medicine used in activating blood to resolve stasis) E and Gushukang group(a medicine used in treating osteoporosis in clinic) F.After nine weeks,we detect the bone density of femur with dual energy X-ray bone density instrument and the expression level of A-FABP in the femur by Western blotting method.We also assay the concentration of high density lipoprotein cholesterol(HDL-C)in serum by automatic biochemistry analyzer.Results:①Contracted with normal group,the density of femur the content of HDL-C in model group is markedly reduced,while the expression level of A-FABP in model group is markedly increased(P〈0.01).②Contracted with model group,the density of femur and the content of HDL-C in tonifying kidney group is markedly increased,while the expression level of A-FABP is markedly content of HDL-C in GIO justify that one molecular mechanism of GIO may be the abnormal lipid metabolism.Conclusion:By means of tonifying kidney,we find out that a prescription containing Cornu Cervi Pantotrichum,Concha Ostreae and Herba Epimedii can prevent and cure GIO by means of regulating lipid metabolism.
