慢性乙型肝炎患者外周血单核细胞亚群表面程序性死亡配体1表达

The expression of the surface of PD-L1 on circulating monocyte subsets in chronic hepatitis B patients

摘要 目的探讨程序性死亡配体1(PD-L1)在慢性乙型肝炎患者外周血单核细胞亚群表面的表达情况。方法利用荧光抗体标记结合多色流式技术,检测慢性乙型肝炎患者外周血单核细胞各亚群表面PD-L1分子的表达。结果慢性乙型肝炎患者外周血单核细胞PD-L1的表达率与血清AST水平呈正相关(r=0.358,P=0.035),且免疫活化(IA,即免疫清除期)患者的CD14highCD16+单核细胞上PD-L1的平均荧光强度(16.75±9.36)最高。结论慢性乙型肝炎患者外周血单核细胞PD-L1表达上调,且与AST水平正相关,可能与乙型肝炎感染慢性化相关。 Objective To investigate the expression of PD-L1 on the surface of circulating monocyte subsets in patients with chronic HBV infection. Methods Immunofluorescent staining and flow cytometry assay were used to measure the expression of PD-L1 on the surface of circulating monocyte subsets from chronic HBV infected patients. Results A significantly positive correlation was found between PD-L1 expression (r=0.358, P=0.035) and serum AST levels, the highest levels of PD-L1 was found on the surface of CD14highCD16+ monocytes(MFI: 16.759.36) from patients in immune active phase. Conclusion PD-L1, which is up-regulated on the surface of monocytes from chronic HBV-infected patients, is positively correlated to serum AST levels, and might play a role in the persistence of HBV infection.
