PKB/CDC25B信号通路对甲状腺鳞癌SW579细胞生长增殖的调控作用

摘要 目的探讨PKB/Cdc25B信号通路对甲状腺鳞癌SW579细胞生长增殖的调控作用。方法体外培养甲状腺鳞癌SW579细胞,实验分为对照组、Wortmannin组和DMSO组,在倒置显微镜下观察细胞生长情况及形态变化;用western blot方法检测PKB、CDC25B蛋白表达和PKB-pS473、CDC25B-pS353及CDC2-pTyr15的磷酸化状态;用ELISA方法检测MPF活性。结果与对照组相比,DMSO组细胞形态无明显变化,各项检测指标无明显差异(P>0.05);Wortmannin组细胞形态回缩,PKB和CDC25B蛋白表达明显下降(P<0.01),PKB-pS473和CDC25B-pS353的磷酸化水平显著下调(P<0.01),而CDC2-pTyr15的磷酸化水平显著上调(P<0.01),MPF活性明显下降(P<0.01)。结论甲状腺鳞癌SW579细胞内源性PKB可以通过调控CDC25B对MPF活性及细胞增殖产生影响。 【Objective】To explore the effect of PKB/Cdc25B signaling pathway on proliferation of thyoid squamous cell carinoma SW579.【Methods】SW579 cells were cultured in vitro and divided into control,wortmannin and DMSO groups randomly.The cells were observed with an inverted microscope.PKB,CDC25B,PKB-pS473,CDC25B-pS353 and CDC2-pTyr15 protein expression levels were determined by Western blot.The activity of MPF was detect by ELISA.【Results】Compared with the control group,DMSO group showed no significant differences in the cell morphology and all detections;While the wortmannin group cell appeared retraction.The protein levels of PKB,CDC25B,PKB-pS473,CDC25B-pS353 obviously reduced while CDC2-pTyr15 increased evidently.The activity of MPF decreased sharply.【Conclusion】Endogeneous PKB could conduct the activity of MPF and Cell proliferation by CDC25B in thyoid squamous cell carinoma SW579.
