摘要
以灭活马流感病毒(EIV)A/Equine/Jilin/1/1989(H3N8)为免疫原,免疫Balb/c小鼠,经常规细胞融合后,用血凝抑制试验(HI)和间接ELISA方法筛选获得3株(3C2、5G10和5A10)能稳定分泌H3N8亚型马流感病毒单克隆抗体(mAb)的杂交瘤细胞株。其中3C2和5G10为IgG2α,5A10为IgM亚类,腹水抗体效价检测显示,3C2和5G10为1∶64 000,5A10为1∶32 000。HI和Western blot结果表明,3株单克隆抗体均为EIV血凝素特异性单克隆抗体。特异性试验结果表明,所获得的mAb不与H7N7亚型马流感病毒、马动脉炎病毒(EAV)、马传染性贫血病毒(EIAV)、马日本脑炎病毒(JEV)发生交叉反应。研究为建立单克隆抗体介导的检测马流感病毒抗原和抗体的血清学方法以及为实现马流感标准化诊断奠定基础。
Three monoclonal antibodies to equine influenza virus were obtained by fusing SP2/0 myeloma cells with spleen cells of Balb/c mice immunized with A/Equine/Jilin/1/1989(H3N8),which were designated as 3C2,5G10 and 5A10 respectively.In which 3C2 and 5G10 as the IgG subclasses,5A10 for the IgM subclass antibody titer detection of ascites,3C2,and 5G10 to 1∶64 000,5A10 is 1∶32 000.Both HI and Western blot results showed that three monoclonal antibodies are EIV hemagglutinin-specific monoclonal antibodies.By hemagglutination inhibition test(HI),indirect ELISA and Western blot assay,these monoclonal antibodies reacted specifically with EIV but not with H7N7 subtype EIV,equine arteritis virus(EAV),equine infectious anemia virus(EIAV),horse encephalitis virus(JEV).The results suggested that these monoclonal antibodies are very useful in diagnosing equine influenza.
