摘要
目的探讨去铁胺(DFO)对人成骨细胞株(hFOB1.19)骨相关基因Ⅰ型胶原(COL1)、骨保护素(OPG)和骨钙素(BGP)基因表达的影响。方法 hFOB 1.19在34℃下进行体外培养,培养基中加入不同浓度的DFO(5、10、20μmol/L)培养48 h后,收集细胞,分别抽取总RNA,采用半定量RT-PCR法检测hFOB 1.19中COL1、OPG、BGP mRNA的表达。结果 DFO浓度依赖性上调hFOB 1.19 COL1、OPG、BGP mRNA的表达(P<0.05)。结论 hFOB 1.19的COL1、OPG、BGP是hFOB 1.19活性相关基因;DFO对hFOB 1.19功能有促进作用,对防止骨质疏松症有一定潜在价值。
Objective To investigate the effects of deferoxamine(DFO) on the gene expressions of osteoprotegerin(OPG),collagen-I(COL1) and osteocalcin(BGP) in human osteoblasts in vitro.Methods Osteoblasts were cultured at 34℃ in vitro.The different DFO concentrations(5,10,20μmol/L) were added into the culture medium.After treated with different concentrations of DFO for 48h,total RNA was isolated from osteoblasts.Expressions of COL1,OPG and BGP were detected by semiquantitative RT-PCR.Results DFO remarkably increased the expressions of COL1,OPG and BGP mRNA in human osteoblasts in a dose-dependent manner.Conclusion DFO upregulates the expressions of COL1,OPG and BGP in osteoblasts and has a potential role in preventing and treating osteoporosis.
