摘要
目的研究表达变异链球菌表面蛋白PAcA、PAcP和霍乱毒素B亚单位(CTB)的转基因番茄防龋疫苗的致突变作用和生殖毒性作用。方法将42只5周龄清洁级昆明小鼠随机分为7组,分别为高(5 g/kg)、低(2 g/kg)剂量阴性对照(正常番茄汁)组和PAcA/CTB、PAcP/CTB蛋白-番茄汁染毒组以及阳性对照(100 mg/kg环磷酰胺)组,每组6只,雌雄各半;除阳性对照组采用腹腔注射方式染毒外,其余6组均采用经口灌胃方式进行染毒,每天1次,连续染毒4 d;测定骨髓细胞微核率。同法进行精子畸形试验(阳性对照为20 mg/kg环磷酰胺)。将60只5~8周龄清洁级SD大鼠随机分为3组,分别为PAcA/CTB、PAcP/CTB蛋白-番茄汁染毒组和对照(非转基因番茄汁)组,每组20只,雌雄各半。进行子一代生殖毒性试验。结果转基因番茄汁染毒小鼠骨髓细胞微核率、精子畸形率与阴性对照组比较,差异均无统计学意义。转基因番茄汁染毒亲代大鼠体重、孕鼠受孕率、正常分娩率与对照组比较,差异均无统计学意义;转基因番茄汁染毒胎鼠出生存活率、哺乳存活率,仔鼠生长发育(体重、身长、尾长)及骨骼发育情况与对照组比较,差异均无统计学意义;病理学检查均未见明显病理改变。整个试验期内,各组亲代、子代大鼠均未出现明显的中毒反应。结论本试验所用的转基因番茄防龋疫苗不会引起致突变作用和生殖毒性作用。
Objective To study the animal safety evaluation on the transgenic tomato anti-caries vaccine which carrying mosaic protein of Streptococcus mutaus surface protein PAc region and cholera toxin B subunit. Methods Forty-two Kunming mice of five weeks old were randomly divided into seven groups, the transgenic tomato group, the non-transgenic tomato group and positive control group,six in each (three female ,three male). Only in positive control group, the treatment was conducted by abdominal injection,in the other groups by gavage, once a day, for four consecutive days. The bone marrow micronuclei incidence rates and teratospermia rate were determined. Sixty SD rats of five to eight weeks old were divided into the transgenic tomato group and the non-transgenic tomato group randomly,20 in each (10 female, 10 male), the male rats were treated at 1 g/kg for eight weeks, the female for two weeks. The generation reproductive toxicity test was conducted. Results No significant difference was observed in bone marrow micronuclei incidence rates and teratospermia rate between experimental groups and the negative group (P〉0.05). No generation reproductive toxicity was observed in the study. Conclusion In the study,the transgenic tomato anticaries vaccine doesn't present obvious mutagenesis and reproductive toxicity.
