电针和穴位埋线对单纯性肥胖大鼠脂质代谢基因PPAR-γ mRNA表达及相关脂代谢酶的影响 被引量:31

摘要 目的:探讨针灸减肥的作用机制。方法:SD雄性大鼠120只,随机选12只作为正常组,其余采用高脂高糖饮食制备单纯肥胖大鼠模型。将造模成功的36只肥胖大鼠随机分为模型组、电针组和埋线组,各12只。电针组和埋线组均取"后三里""天枢""脾俞"穴,分别给予电针和穴位埋线干预,电针每天1次,埋线7 d 1次。15 d后,观察各组大鼠体质量变化,检测各组大鼠血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)水平,检测肝脏脂蛋白脂酶(LPL)和肝脂酶(HL)活性,脂肪组织过氧化物酶体增殖物活化受体γ(PPAR-γ)mRNA表达。结果:干预后,电针组和埋线组体质量及增加体质量均低于模型组(均P<0.01)。与正常组比较,模型组血清TC、LDL-C水平显著升高(均P<0.01);肝脏LPL、HL活性下降(P<0.05,P<0.01),脂肪组织PPAR-γmRNA表达水平减弱(P<0.05)。与模型组比较,电针组和埋线组血清TC均下降(P<0.05,P<0.01),LDL-C水平亦下降(P<0.01,P<0.05),TG水平3组间差异无统计学意义(P>0.05),肝脏LPL活性有所升高(P<0.01,P<0.05),HL活性亦升高(均P<0.01),脂肪PPAR-γmRNA表达水平升高(均P<0.01)。结论:电针和穴位埋线可通过提高脂肪PPAR-γmRNA的表达,增强肝脏LPL和HL活性,降低血清TC和LDL-C水平,从而达到减肥和调节脂质代谢紊乱的目的。 Objective To study the mechanism of electroacupuncture and acupoints catgut-embedding in losing weight. Methods Simple obesity was induced in male SD rats with high-fat and high-glucose diet for 10 weeks. Thirty-six rats with obesity were randomly divided into model-control group, electroacupuncture (EA) group and catgut-embed(ling (CE) group, with 12 rats in each group. Another 12 normal SD rats serving as controls (normal control group) were fed by standard rat chow. Rats in the EA group were treated with electroacupuncture at bilateral acupoints of "Housanli"(ST 36), "Tianshu"(ST 25), and "Pishu"(BL 20), once daily for consecutive 15 days. Rats in the CE group were treated with catgut-embedding at the same acupoints with the EA group, once every 7 days. Rats in the other two groups were not interfered. After 15 days of treatment, changes of body weight were observed, the levels of total cholesterol(TC), triglyceride(TG) and low-density lipoprotein eholesterol(LDL-C) in serum were assayed, the activities of lipoprotein lipase (LPL) and hepatic lipase (HID in liver tissues were measured, and the mRNA expression levels of Peroxisome proliferators activated receptor-g(PPAR-y) in adipose tissues were determined. Results After the intervention, body weight and increase of body weight in the EA and CE groups were significantly lower than those in the model-control group(all P〈0.01). Compared with the normal control group, there was significant increasing of levels of TC and LDI.-C in serum(both P〈 0.01), descending activities of LPL and HL in liver tissues(P〈0.05,P〈0.01), and decreased expression levels of PPAR-7 mRNA in adipose tissues in the model-control group (P〈O. 05). Compared with the model-control group, there were obviously decreased in TC, LDL-C Level in serum in EA and CE groups(P〈0.05 ,P〈0.01), while there was no difference in TG levels among three groups (P〉0.05). The activities of LPL and HL were also significantly enhanced in EA and CE groups as compared with those in the model-control group (P〈0.01 ,P〈0.05), so were the expression levels of PPAR-γ mRNA in adipose tissues (both P〈0.01). Conclusion Electroacupuncture and acupoint catgut-embedding can regulate the lipid metabolic disorders in rats with obesity through enhancing the expression of PPAR-γ mRNA in adipose tissues, activating the activities of LPL and HL and decreasing the levels of TC and LDL-C in serum.
