摘要
目的采用HPLC法建立南五味子和五味子药材的指纹图谱,并同时测定4种木脂素。方法以五味子酯甲、五味子甲素、五味子醇甲和五味子乙素为对照,采用Eclipse XDB-C18色谱柱(150 mm×4.6 mm,5μm),乙腈-四氢呋喃-水梯度洗脱;检测波长220 nm;体积流量1.0 mL/min进行试验,用中国药典委员会颁布的中药色谱指纹图谱相似度评价系统软件(版本:2004AB)计算处理,分别建立南五味子、五味子甲醇提取物的指纹图谱,并同时测定4种木脂素。结果南五味子药材指纹图谱中标定了25个共有峰,五味子药材指纹图谱中标定了24个共有峰;不同产地南五味子指纹图谱相似度均大于0.9,但其主要木脂素成分量差异较大;不同产地五味子指纹图谱相似度亦大于0.9,但南五味子、五味子指纹图谱相似度小于0.3。结论南五味子、五味子药材的指纹图谱特征性和专属性强,结合木脂素成分的测定,可为全面控制药材的质量提供科学方法。
AIM To establish an HPLC fingerprint spectrum and determine four lignans of Schisandrae sphenantherae Fructus and Schisandrae chinensis Fructus.METHODS Deoxyschizandrin,schisantherrin A,schisandrin and γ-schisandrin served as the reference substances.The assay was conducted on Eclipse XDB-C18 column(150 mm×4.6 mm,5 μm) with acetonitrile-tetrahydrofuran-water as mobile phase.The flow rate was 1.0 mL/min and the detection wavelength was set at 220 nm.Similarity evaluation system for chromatographic fingerprint of TCM(2004 AB) was used in data analysis.RESULTS Twenty-five common peaks were pointed out from the HPLC fingerprint of Schisandrae sphenantherae Fructus and 24 common peaks from Schisandrae chinensis Fructus;Schisandrae sphenantherae Fructus from different habitats were of strong similarity in HPLC fingerprint chromatogram,but there were obvious differences in contents of main lignans;Schisandrae chinensis Fructus in different batches were of strong similarity,but both similarities were less than 0.30.CONCLUSION This method is simple and reliable.The HPLC fingerprint and the determination of the main lignans can be used for their quality control.
