摘要
目的探索HMBA及其衍生物HMBA-Ⅰ和HMBA-Ⅱ对人红白血病细胞系K562的诱导分化作用。方法细胞生长曲线绘制、集落形成试验,检测HMBA及其衍生物对细胞生长的影响,以Westernblot检测p21、c-fos蛋白表达,以联苯胺染色显示血红蛋白。结果HMBA-Ⅰ,HMBA-Ⅱ呈现对K562细胞生长有不同程度的抑制作用,基因表达产物血红蛋白联苯胺染色呈阳性反应,p21、c-fos蛋白表达有不同程度的增高。结论HMBA-Ⅰ,HMBA-Ⅱ对K562有一定的诱导分化活性,作用机制可能与p21、c-fos表达增高有关。
Objective\ To Study the differnetiation inducing effect of HMBA and its derivatives HMBA Ⅰ and HMBA Ⅱ on K562 cells. Methods\ K562 cells were cultured and treated with HMBA and its derivatives. We studied their effect of inducing differentiation on K562 cells by benzidine staining and other methods often used. The c fos, p21 gene expression were detected by Western blot, and cell cycle analyzed. Result\ It was revealed that both HMBA Ⅰ,HMBA Ⅱ as well as HMBA exerted growth inhibition effect on K562 cells and the expression of p21 and c fos proteins were increased. Conclusion\ HMBA Ⅰ and HMBA Ⅱ as well as HBMA are potential cell differentiation inducers.
