摘要
通过紫外诱变和微波辐射米曲霉菌株,筛选出高产纤维素酶和植酸酶米曲霉菌株ZQH48,并进行了固体发酵条件的优化.优化后的培养基配方为:菌渣:麸皮=1∶1,氯化铵12%,蛋白胨2%,硫酸镁0.07%,氯化钠0.5%,磷酸二氢钾0.02%,固水比为1∶1;最佳培养温度35℃.优化后纤维素酶活力可达161.55 U/g,较原始菌株提高5.42倍;植酸酶活力达1 541.13 U/g,较原始菌株提高1.56倍.
The strain of Aspergillus Oryzae ZQH48 overproducing cellulase and phytase was isolated by UV and microwave radiation,at the same time,the cultural factors and conditions of solid-state fermentation were also optimized.The optimal culture condition was 12%ammonium chloride,2%peptone,0.07%magnesium sulfate,0.5%sodium chloride,0.02%potassium dihydrogen phosphate,ratio of liquid to material and mushroom dregs to bran 1∶1,35℃.In this condition,the activity of cellulase and phytase of the ZQH48 strain increased by 161.55 U/g and 1 541.13 U/g respectively,which was 5.42 times and 1.56 times higher than the parent strain.
