促红细胞生成素对毛果芸香碱诱导的小鼠癫痫发作脑保护的研究 被引量:3

摘要 目的研究促红细胞生成素(EPO)对毛果芸香碱诱导的小鼠癫痫发作海马神经元细胞的保护作用。方法用毛果芸香碱350mg/kg腹腔注射C57/B6小鼠诱导癫痫发作,予癫痫发作2h后用地西泮1mg/kg终止其发作。EPO组予毛果芸香碱诱发癫痫前2 h、癫痫发作后2及24 h腹腔注射EPO(50 g/kg),对照组于相同时间点注射等体积0.9%氯化钠注射液,观察各组癫痫发作后病死率及诱发癫痫发作潜伏期时间。应用NeuN免疫组化染色,观察各组海马神经元脱失情况,并与正常小鼠进行比较。结果 (1)EPO组的癫痫发作后病死率与对照组差异无统计学意义(>0.05),但EPO组癫痫发作潜伏期时间明显长于对照组(<0.05)。(2)免疫组化显示:EPO组NeuN阳性细胞数明显低于正常组,但明显高于对照组(<0.01)。结论 EPO可以减轻小鼠癫痫的临床发作,减轻小鼠海马神经损伤,抑制神经元细胞凋亡,从而发挥其神经保护作用。 Objective To explore the protective effect of erythropoietin on damaged neurons in the hippocampus of C57/B6 mice with epileptic seizure.Methods The status convulsion model was produced by pilocarpine,350mg/kg,in-traperitoneal injection.2 hours later,convulsion were stopped by diazepam.For the EPO group,pilocarpine was given 2 hours before convulsion,2 hours after convulsion and 24 hours after convulsion,by intraperitoneal injection(50 g/ kg);0.9% sodium chloride injection group was given with the isometric 0.9% sodium chloride injection.Mortality and incubation time were observed.The brain tissues were used to make the paraffin sections for NeuN straining in the hippocampal CA1,CA2,CA3,CA4 regions by the method of immunohisochemistry.Results There was no sig-nificant difference on mortality between EPO cure group and 0.9% sodium chloride injection group(0.05),but the incubation was longer(0.05).The NeuN immunohistochemistry showed that the neurons loss in hippocampus in EPO group was significantly less than that in saline group.Conclusions EPO can reduce the neurons apoptosis in the hippocampal after status convulsion in C57/B6 mice.EPO may have the anti-apoptosis effect,and then play a part in nueroprotection.
