摘要
选用初始体重为(39.68±3.05)g的草鱼作为试验对象,在全植物蛋白基础饲料中,设5个不同的植酸酶添加水平(500、750、1 000、1 250、2 500 U/kg),另设2个对照组,对照组1(D1):全植物蛋白基础饲料添加无机磷(Ca(H2PO4)2),对照组2(D2):全植物蛋白基础饲料不添加植酸酶和无机磷,开展了为期8周的饲养试验。研究了植酸酶对草鱼生长、非特异免疫及消化酶活力的影响。结果表明:对照组1鱼体的特定生长率最大,FCR显著低于其它各组(P〈0.05);与对照组2相比,当植酸酶添加水平大于1 000 U/kg时,草鱼的饲料系数显著降低、蛋白质效率、特定生长率得到显著提高(P〈0.05)。当植酸酶添加水平大于1 000 U/kg时,血清超氧化物歧化酶(SOD)活性显著大于对照组2(P〈0.05),对照组1的血清SOD活性显著高于其它各组(P〈0.05);血清及肝胰脏中各组碱性磷酸酶(AKP)活性无显著性差异(P〉0.05);添加植酸酶后肝胰脏中SOD活性增强,且当植酸酶含量为1 250 U/kg时,肝胰脏SOD活性显著高于对照组1和对照组2(P〈0.05)。当植酸酶添加水平大于1 000 U/kg时,草鱼肝胰脏和肠道中蛋白酶、淀粉酶和脂肪酶活性都明显增强。结果得出,当植酸酶添加水平为1 000~1 250U/kg时,全植物蛋白饲料中添加植酸酶有助于草鱼的生长、非特异性免疫机能的提高,并能有效增强草鱼肝胰脏和肠道中的消化酶活性。
A 8-weeks laboratory trial was designed to study the effect of phytase on growth,non-specific immunity and the digestive enzyme activity of juvenile grass carp(Ctenopharyngodon idellus) fed plant protein based diet.Seven trial diets were formulated to feed fish(with mean initial body weight of(39.68±3.05 g)) that were divided into 5 experimental groups and 2 control groups.Five triplicate experimental groups were supplemented with five different levels of phytase(500,750,1 000,1 250,2 500 U/kg).The positive control group(D1) was supplemented with 1.5% monobasic calcium phosphate(Ca(H2PO4)2);and the negative control group(D2) was not supplemented with phytase and monobasic calcium phosphate.The trial was conducted in tanks indoor with stocking density of 15 fishes each tank.The results showed that fish in positive control group had the best specific growth rate and lowest feed conversion rate(FCR),significantly different from other groups(P0.05);the FCR of experimental group 3(1 000 U/kg) was significantly lower,and protein efficiency ratio(PER) was significantly higher than that of negative control group(P0.05).For the non-specific immunity of juvenile grass carp,the serum superoxide dismutase(SOD) activity of grass carp in(D1) was significantly higher than that of other groups(P0.05),whereas,when the phytase cotent was higher than 1 000 U/kg,the serum SOD activity of grass carp was higher than that of(D2)(P0.05).The kaline phosphatase(AKP) activities had no significant differences in serum and hepatopancreas of different groups(P0.05).The SOD activity in hepatopancreas was improved to some extent after supplemented with phytase especially,when the content of phytase was 1 250 U/kg(P0.05).The activities of digestive enzymes(protease,lipase and amylase) in hepatopancreas and intestine could be improved when the level of phytase was higher than 1 000 U/kg.The results concluded that the phytase had a capacity to improve the growth,activity of digestive enzymes in hepatopancreas and intestine,and had a positive effect on the non-specific immunity of juvenile grass carp when the content of phytae was 1 000-1 250 U/kg.
