摘要
用TTC法测定管花肉苁蓉种子活力,结果表明,将剥去种皮的种子用0.5%的TTC溶液在37℃下染色48h以上可以获得稳定的活力测定结果,而未剥种皮染色120h仍不能测定活力;花序上部粒径〉0.5mm的种子活力为10%-24%;同一花序下部相同粒径的种子活力约为40%;花序下部粒径〈0.5mm的种子活力〈1%;套袋收获的种子活力明显高于直接采收花序获得的种子。
In this study, Tetrazolium was used to assess the viability of Cistanche tubulosa seeds which were treated or collected in different ways. Results showed that seeds stripped of coats were stained in 0.5% TTC solution at 37 ℃ and the percentage of stained seeds wasn' t increase when seeds were stained more than 48 h, while few seeds with coats were stained even for 120 h in the same conditions. Seed( 〉0.5 mm in diameter) viability was 10% -24% in upper positions of inflorescences but lower positions in the same inflorescences produced abundant viable seeds (approximately 40% ). Seed( 〈0.5 mm in diameter) viability was nearly 1% in lower positions of inflorescences. Seeds collected by bagging method had higher viability than that of seeds collected from the inflorescence directly.
