摘要
目的:建立同时测定杜仲雄花中京尼平苷酸、绿原酸、栀子苷3种成分含量的方法。方法:采用双波长高效液相色谱法。色谱柱为Agilent Zorbax Extend C18(250mm×4.6mm,5μm),流动相为甲醇-水-醋酸(15∶85∶1),检测波长为238nm(测定京尼平苷酸和栀子苷)和327nm(测定绿原酸),流速为1mL·min-1,柱温为30℃。结果:京尼平苷酸、绿原酸、栀子苷的进样量线性范围分别为0.0892~5.3520μg(r=0.9995)、0.1052~6.3120μg(r=0.9995)、0.1024~6.1440μg(r=0.9995);三者平均加样回收率分别为99.24%、98.86、101.44%,RSD分别为2.17%、1.95%、2.08%(n=6)。结论:双波长或多波长法能方便、快速地对中药中多种成分同时测定,能满足中药多成分质量控制的要求。
OBJECTIVE:To develop the method for the content determination of geniposidic acid,chlorogenic acid and geniposide in male flowers of Eucommia ulmoides.METHODS:Dual-wavelength HPLC was adopted.The analytical column was Agilent Zorbax Extend C18(250 mm×4.6 mm,5 μm) column with mobile phase consisted of methanol-water-acetic acid(15 :85 :1) at flow rate of 1 mL·min-1.The detection wavelength was set at 238 nm for geniposidic acid and geniposide,and at 327 nm for chlorogenic acid.The column temperature was 30 ℃.RESULTS:The linear ranges of geniposidic acid,chlorogenic acid and geniposide were 0.089 2~5.352 0 μg(r=0.999 5),0.105 2~6.312 0 μg(r=0.999 5),0.102 4~6.144 0 μg(r=0.999 5).Average recoveries of them were 99.24%,98.86%,101.44% and RSDs were 2.17%,1.95%,2.08%,respectively(n=6).CONCLUSION:Dual or more wavelength HPLC method can be used for simultaneous determination of ingredients of traditional Chinese medicine,and can satisfy the requirements of quality control.
