摘要
目的:探讨1,6-二磷酸果糖(FDP)对脑白质损害(WMD)大鼠皮质生长相关蛋白43(growthassociation protein43,GAP-43)表达的影响。方法:选用2日龄新生大鼠50只,随机分为对照组(12只)、FDP组(13只)、生理盐水组(12只)及WMD组(13只),后3组通过结扎右侧颈总动脉,吸入氧体积浓度为6%的氮氧混合气,制作新生大鼠WMD模型,分别进行相应的实验干预。损伤后第7天和14天时将大鼠处死,取脑组织应用Western Blots方法检测皮质GAP-43的表达,损伤后30d进行感觉运动功能检测,以观察治疗效果。结果:FDP组感觉运动功能较WMD组和生理盐水组有明显改善(P<0.01或P<0.05)。WMD组、生理盐水组和FDP组皮质GAP-43的表达在干预后7d时高于对照组,在14d时FDP组高于WMD组、生理盐组和对照组(P<0.01)。结论:FDP可以上调WMD大鼠皮质GAP-43表达,改善WMD大鼠的脑功能。
Objective To study the effects of fructose-1,6 diphosphate (FDP) on the expression of growth association protein 43 (GAP-43) in the cortex of white matter damage (WMD) in neonatal rats. Methods Two-day-old Sprague-Dawley rats (n = 50) were randomly assigned into normal group (n = 12), FDP group (n = 13), saline group (n = 12) and WMD group (n = 13). Firstly, the latter 3 groups were produced WMD models by dissecting and ligating the fight common carotid artery, and by exposing to 6% oxygen and -94% nitrogen gas mixture for 4 h. And then, they received the intervention study, respectively. The abilities of sensorimotor movement (hanging test and inclined plane test) were detected on 30 days after damage, and western blot was used to examine the expression of GAP-43 in cortex on the 7th day and 14th day after damage. Results The sensorimotor movement abilities of FDP group were much higher than those of WMD group and saline group (P 〈 0.01 or P 〈 0.05). Compared with that of the control group, the expressions of GAP-43 in the FDP group, saline group and WMD group were higher on the 7th day after damage (P 〈 0.01 ). The expression of GAP-43 was up-regulated on the 14th day in the FDP group (P 〈 0.01 ). Conclusion FDP can increase the expression of GAP-43 in the cortex and improve the sensorimotor movement ability of WMD neonatal rat.
