摘要
目的研究阴沟肠杆菌Ι类整合子可变区基因盒的种类及位置,探讨Ι类整合子携带的耐药基因盒特点,分析耐药基因盒与阴沟肠杆菌耐药性的关系。方法采用纸片扩散法(K-B法)测定抗生素的敏感性;PCR扩增阴沟肠杆菌Ι类整合子阳性株中的可变区基因盒,并测序分析。结果可变区一共扩增出15种基因盒,其中耐药基因盒7种:aadA2-dfrA12、aadA2-aadB、aadA2、aadB、dfrA12、aac(6')-Iica-catB3和blaIMP-4,以aadA2-dfrA12(1913bp)最常见。许多质粒DNA和染色体DNA的PCR扩增产物相同。结论Ι类整合子可变区携带的耐药基因盒以耐氨基糖苷类和耐甲氧苄氨嘧啶类抗生素基因为主。耐药基因在阴沟肠杆菌中存在水平播散和垂直传播的现象。
Objective To investigate the types and distribution of the genes cassettes in variable region of class I integron in Enterobacter cloacae, analysis the feature of resistance genes cassettes and relationships between resistance genes cassettes of class I integron and Enterobacter cloacae resistance. Methods Antibiotics susceptibility test was used by disk diffusion method( K-B method); detecting genes cassettes in variable region of class Iintegron using PCR and done sequence analysiss. Results 15 gene cassettes were amplified in the variable region of class Iintegron, including 7 resistant gene cassettes: aadA2-dfrA 12, aadA2-aadB, aadA2, aadB, dfrA 12, aac(6')-Iica-catB3 and btaIMP-4. The aadA2-dfrA 12 was the most prevalent in variable region of class Iintegron. The sizes of amplification products of many plasmids and chromosomes DNA are same. Conclusion There were mainly aminoglycosides and trimethoprim antimicrobial resistant genes cassettes in the variable region of class I integron. Antimicrobial resistant genes can be disseminated by horizontal and vertical trsmission in Enterobacter cloacae.
