摘要
目的利用去甲基化酶5-氮-2’-脱氧胞苷(5-aza-CDR)对脾酪氨酸激酶(Syk)基因激活的作用,观察Syk基因的激活对人胃癌SGC-7901细胞血管内皮生长因子(VEGF)表达的影响,从而研究Syk基因的激活对人胃癌SGC-7901细胞生长和转移的影响。方法检测用5-aza-CDR处理后的人胃癌SGC7901细胞株的生长情况、SYK基因的表达水平及血管内皮生长因子(VEGF)mRNA的表达水平,与未接触5-aza-CDR的对照组进行对比。结果经5-aza-CDR处理过的人胃癌SGC-7901细胞株的生长明显受抑;该细胞株SYK基因的表达水平较对照组明显升高,血管内皮生长因子(VEGF)mRNA的表达水平较对照组明显减弱(P<0.05)。结论 Syk基因的表达能够抑制人胃癌SGC-7901细胞株生长,抑制该细胞株VEGF的表达。血管内皮生长因子(VEGF)的表达抑制可能是SYK基因的激活,从而抑制该细胞株的生长和转移的机制之一。
Objective Demethylase 5-N-2 '-deoxycytidine(5-aza-CDR) was used to activate the spleen tyrosine kinase(Syk) gene,to observe the effect of Syk gene activation on vascular endothelial growth factor(VEGF) expression in human gastric cancer cell line SGC-7901 and study the effect of the activation of Syk gene on growth and metastasis in it.Methods 5-aza-CDR was used to incubate with human gastric cancer cell line SGC7901,the cell growth state、Syk gene expression and VEGF mRNA expression were detected,to compare with the control group non-contact 5-aza-CDR.Results The growth of human gastric cancer cell line SGC7901 was significantly inhibited after incubated with 5-aza-CDR;The cell lines SYK gene expression was significantly higher than the control group,vascular endothelial growth factor(VEGF) mRNA expression were significantly reduced compared with the control group.(x2=,P0.05).Conclusion Syk gene re-expression could inhibit the growth of human gastric cancer cell line SGC-7901 and the expression of VEGF in the cell lines,inhibition the expression of vascular endothelial growth factor(VEGF) may be one of the mechanisms that activated SYK gene inhibited the growth and metastasis of cell lines.
