转BADH-反义4CL基因二色胡枝子的组培快繁技术 被引量:2

On rapid propagation by tissue culture and transplanting techniques of the transgenic Lespedeza bicolor

摘要 为探索建立转BADH-反义4CL基因胡枝子组培快繁体系和掌握试管苗移栽技术,以转基因和非转基因二色胡枝子组培苗为材料,对植株进行茎段增殖、茎段生根的快繁研究,同时进行基质、光照、苗质量对试管苗移栽成活率影响研究。结果表明,非转基因植株茎段增殖最佳培养基配方为:1/2MS+0.5 mg/L 6-BA+0.1 mg/L NAA+0.2 mg/L IBA,增殖系数4.46;转基因植株茎段增殖最佳培养基为1/2MS+1.0 mg/L 6-BA0.1 mg/LNAA0.3 mg/L IBA,增殖系数3.95。非转基因植株茎段生根最佳培养基为1/2MS+0.4 mg/L IBA,生根率为100%;转基因植株茎段生根培养基:1/2MS+0.2 mg/L NAA+0.4 mg/L IBA,生根率为99%。非转基因植株和转基因植株的移栽基质为草炭∶蛭石∶珍珠岩(体积比1∶1∶1/4)、遮一层遮阴网、苗质量为正常苗时移栽成活率较高。 In order to develop a propagation technique of tissue culture and gain transplanting techniques for test-tube seedlings of the non-transgenic and transgenic Lespedeza bicolor,the stem segment proliferation,rooting and the effects of some factors on survival rate were studied.The factors included lighting,medium and seedling quality.The results showed that the proliferation medium of the non-transgenic Lespedeza bicolor was 1/2MS+0.5 mg/L 6-BA+0.1 mg/L NAA+ 0.2 mg/L IBA and the coefficient of multiplication could reach up to 4.46.The proliferation medium of transgenic plants was 1/2MS+1.0 mg/L 6-BA+0.1 mg/L NAA+ 0.3 mg/L IBA and the coefficient of multiplication was 3.95.The rooting medium of non-transgenic Lespedeza bicolor was 1/2MS+0.5 mg/L 6-BA+0.1 mg/L NAA+ 0.2 mg/L IBA and the rooting ratio could reach up to 100%.For the transgenic plants,the rooting medium was 1/2MS+0.2 mg/L NAA+ 0.4 mg/L IBA and the rooting ratio was 99%.The non-transgenic and transgenic Lespedeza bicolor were higher surviving rate when transplanted to the disinfected composing soil(turf∶vermiculite∶perlite,1∶1∶1),covered a layer of shate net,seedling quality was nomal seeding.
