摘要
目的探索流感病毒在MDCK细胞中培养的最适条件。方法将流感病毒分别以直接接种法和吸附接种法接种于MDCK细胞,比较其接种效果;选择其中更合理的方法,对其培养条件(培养时间、血清浓度、胰酶浓度、接种量和传代次数)进行比较和优化。结果直接接种法相对于吸附接种法更具优势。最佳收获病毒时间为72 h;胎牛血清对病毒的增殖有明显抑制作用;终浓度为2.5μg/ml的胰酶可明显提高细胞增殖病毒的血凝效价。结论通过对MDCK细胞培养流感病毒条件的优化,可以明显提高流感病毒培养效率。
Objective To explore the most suitable condition for culturing influenza virus on MDCK cells.Methods Influenza viruses in different concentrations were propagated on MDCK cells,and then underwent serial subcultures.The viruses were cultured in the maintenance solution with FBS or pancreas enzyme in different concentrations and then harvested on different time points.The corresponding haemagglutinin titers(HA) were detected.Results It was observed that the proliferation of influenza virus was inhibited in the maintenance solution containing FBS,but significantly increased in maintenance solution containing pancreas enzyme with the final concentration of 2.5 μg/ml.The best harvest time was 72 hrs.Conclusions An optimized condition of propagating influenza virus on MDCK cells is successfully established,which can be used to propagate influenza virus effectively.
