摘要
目的:建立单侧肾梗阻(unilateral ureteral obstruction,UUO)致肾纤维化模型,探讨真武汤对肾纤维化大鼠24h尿量、尿蛋白含量、血液生化等指标的影响。方法:将大鼠随机分为5组,除正常对照组外,大鼠采用结扎左侧输尿管的方法建立肾纤维化模型。造模大鼠随机分为4组:模型组、氯沙坦钾组(20 mg/kg.d)、真武汤治疗组(2.1、9.8 g生药.kg-1.d-1),检测0、14、28 d大鼠24 h尿量及其尿蛋白含量。给药结束后,取双侧肾,比较肾脏组织形态变化,并检测大鼠的两肾比重及14、28 d大鼠的血液生化指标。结果:真武汤可减少UUO大鼠的24 h尿量,降低其尿蛋白含量,调节肾纤维化大鼠血浆中肌酐及尿素氮水平,对白蛋白、总胆固醇、甘油三酯、低密度及高密度脂蛋白含量影响不明显。结论:真武汤对肾纤维化大鼠蛋白尿及肾功能具有正相调节作用,对大鼠体内脂质代谢也具有一定的影响。
Objective: Investigate the influence of zhen wu decoction on the urine volume of 24 hours,the level of urinary protein and blood biochemical index of renal fibrosis rat induced with unilateral ureteral obstruction.Methods: Besides normal control group,the renal fibrosis rats were divided into 4 groups after ligating the left ureter : model group,UUO group treated by losartan potassium(20mg/kg·d),UUO group treated by zhen wu decoction of low dose(21 original medicinal per kilogram everyday) and high dose(98g original medicinal per kilogram everyday).Detecting the urine volume of 24 hours and the level of urinary protein in 0,14,28 days.Observing the appearance of kidney,measuring it's proportion of body weight and detecting the blood biochemical index in 14,28 days when the rats were killed after 28 days.Result: Zhen wu decoction has effect on regulating the urine volume of 24 hours,reducing the level of urinary protein,alleviating the increasing tendency of creatinine and urea but not on adjusting the level of ALB,TG,TC,LDL and HDL in plasma of renal fibrosis rats.Conclusion: Zhen wu decoction has a positive effect in the aspects of proteinuria and renal function,meanwhile,it can work on lipid metabolism of renal fibrosis rats.
