摘要
根据GenBank中犬瘟热病毒Onderstepoort株序列设计特异性引物,以犬瘟热病毒贵州分离株(CDV-GZ1)RNA为模板,应用RT-PCR对病毒H蛋白编码基因进行了扩增、克隆及序列分析。测序结果表明,CDV-GZ1 H基因ORF由1 824 bp组成,可编码607个氨基酸;与已发表23株其它CDV H基因核苷酸序列相似性在91.1%~99.6%之间,推导氨基酸序列同源性在91.0%~99.2%之间。GZ1株H蛋白含有5个潜在的N-联糖基化位点,与其他毒株相比,N-联糖基化位点的数目与位置均存在差异。系统进化树分析表明,CDV-GZ1株与德国Snyder Hill株和国内广州分离株(GZ0802)具有较高的同源性,推测这些毒株间可能具有更近的亲缘关系。
Two pairs of primers were designed and synthesized based on the sequence of the Onderstepoort strain of canine distemper virus(CDV) in GenBank,total RNA isolated from Vero cells infected with Guizhou isolate of CDV(CDV-GZ1) as templates,the H gene was amplified by reverse transcription polymerase chain reaction(RT-PCR).The amplified fragment was cloned and analyzed.Sequencing results indicated that the H gene section of a 1 824 bp fragment encoding 607 amino acids.Compared with other CDV strains,the nucleotide sequence similarity was among 91.1%-99.6%.The amino acid homology was among 91.4%-99.2%.CDV-GZ1 H gene contained 5 potential N-glycosylation locations based on the derived amino acid sequences,and there were differences in the number and locations of N-glycosylation compared with other CDV strains.Phylogenetic tree analysis results showed that the isolated strain was closely related to the Snyder Hill strain and GZ0802 strain.Therefore,they might have common ancestor.
